高粱Pht1基因家族的鑒定及其進化和表達分析

【摘要】:磷是生物大分子的重要組成成分,也是植物生長所必需的重要營養(yǎng)元素。利用Phytozome數據庫,通過擬南芥Pht1家族、水稻OsPht1家族基因序列在高粱全基因組中的同源比對,鑒定并獲得了11個高粱Pht1家族成員的基因序列、基因結構、蛋白序列和染色體定位等信息。利用熒光定量的方法,檢測了11個SbPht1基因在缺磷和高磷處理下的表達差異。結果表明,在缺磷條件下,大部分SbPht1基因表達量上升。高磷脅迫下,SbPT1、SbPT4、SbPT6、SbPT7、SbPT8、SbPT9和SbPT10在根中的表達量上升,SbPT3和SbPT5在地上部的表達量上升。分析高粱全基因組磷轉運蛋白,對于研究植物在磷吸收過程中的分子生物學基礎,改良植物吸收和利用磷素意義重大。
[Abstract]:Phosphorus is not only an important component of biological macromolecules, but also an important nutrient element for plant growth. Using Phytozome database, the gene sequences, gene structure, protein sequence and chromosome location of 11 sorghum Pht1 family members were identified and obtained by homologous alignment of Arabidopsis Pht1 family and rice OsPht1 family gene sequence in sorghum genome. The expression difference of 11 SbPht1 genes under phosphorus deficiency and high phosphorus treatment was detected by fluorescence quantitative method. The results showed that the expression of most SbPht1 genes increased under the condition of phosphorus deficiency. Under high phosphorus stress, the expression of SbPT1,SbPT4,SbPT6,SbPT7,SbPT8,SbPT9 and SbPT10 in roots increased, while the expression of SbPT3 and SbPT5 increased in shoot. It is of great significance to analyze the whole genome phosphorus transporter of sorghum in order to study the molecular biological basis of phosphorus absorption in plants and to improve the absorption and utilization of phosphorus in plants.
