【摘要】：在進(jìn)行馬尾松抗蟲(chóng)材料轉(zhuǎn)錄組測(cè)序分析時(shí)發(fā)現(xiàn)一個(gè)編碼CYP77A1的cDNA,命名為PmCYP77A1。序列分析結(jié)果顯示,該基因全長(zhǎng)1 620 bp,編碼539個(gè)氨基酸。氨基酸序列分析表明其具有CYP450家族基因的保守結(jié)構(gòu)域,但CYP77A1基因中的FXXGXXXCXG保守特征序列最后一個(gè)堿基被A堿基所代替。與其他植物核酸序列進(jìn)行比對(duì)覆蓋率和同源性均較差,而氨基酸序列比對(duì)覆蓋率達(dá)到88%~96%,但同源性不高,基本都在50%~55%之間,PmCYP77A1與白云杉遺傳聚類最近,與其他被子植物遺傳聚類較遠(yuǎn)。利用熒光定量PCR技術(shù)分析得知,PmCYP77A1在馬尾松的不同組織中均有表達(dá),其中在嫩莖、雌雄花和嫩葉中的表達(dá)量較高,而在球果、成熟組織和根系中的表達(dá)量最低。在不同抗蟲(chóng)材料日變化中PmCYP77A1表達(dá)量出現(xiàn)先升高后降低的表達(dá)趨勢(shì),該基因在抗蟲(chóng)材料中表達(dá)啟動(dòng)早。研究結(jié)果將為進(jìn)一步研究PmCYP77A1的生物學(xué)功能及在馬尾松抗蟲(chóng)防御中的作用機(jī)理奠定基礎(chǔ)。
