【摘要】：研究以油松(Pinus tabulaeformis)的優(yōu)良外生菌根真菌-淺黃根須腹菌(Rhizopogon luteolus)菌絲體為對(duì)象,采用簡(jiǎn)并PCR和末端克隆技術(shù)分離了其磷酸鹽轉(zhuǎn)運(yùn)蛋白基因(RlPT),所分離的RlPT cDNA全長(zhǎng)序列2017 bp,包含一個(gè)1 653 bp的開(kāi)放閱讀框(open reading frame,ORF),編碼550個(gè)氨基酸;基因組DNA長(zhǎng)度約為2 324 bp,包含11外顯子和10個(gè)內(nèi)含子,保守結(jié)構(gòu)域搜索顯示屬于MFS超基因家族。結(jié)果表明:蛋白序列的系統(tǒng)進(jìn)化樹(shù)顯示與淺黃根須腹菌(Rhizopogon luteolus)的磷酸鹽轉(zhuǎn)運(yùn)蛋白的親緣關(guān)系最近。qPCR分析顯示在磷饑餓時(shí),該基因表達(dá)顯著提高。
[Abstract]:The phosphate transporter gene (RlPT), was isolated from the mycelium of (Rhizopogon luteolus), a fine exogenous mycorrhizal fungus of Pinus tabulaeformis (Pinus tabulaeformis) by degenerate PCR and terminal cloning techniques. The isolated RlPT cDNA sequence 2017 bp, contained an open reading frame (ORF) (open reading frame,ORF of 1653 bp, encoding 550 amino acids. The length of genomic DNA was about 2 324 bp, which contained 11 exons and 10 introns. The conserved domain search showed that it belonged to the MFS supergene family. The results showed that the phylogenetic tree of the protein sequence showed the closest phylogenetic relationship with the phosphate transporter of (Rhizopogon luteolus), and the expression of the gene increased significantly during phosphorus starvation.
