富集優(yōu)勢基因型的后備親本篩選以及相關(guān)分子標(biāo)記的遺傳效應(yīng)分析

【摘要】:選取黃河鯉新品系親本和子代共450尾,5對微衛(wèi)星引物和4個基因區(qū)段擴(kuò)增引物開展基因型檢測,并檢測它們與體重的關(guān)聯(lián)性以及分析相應(yīng)的遺傳效應(yīng)。結(jié)果顯示:Koi42和4個SNPs位點(diǎn)對體重有顯著的影響,獲得具有超過富集2個優(yōu)勢基因型的候選親本共13尾,富集優(yōu)勢分子標(biāo)記基因型的候選親本生長性能優(yōu)勢明顯。通過多元逐步回歸分析,利用AIC最佳模型篩選到COⅠ626、D-Loop253和Koi42共3個位點(diǎn),發(fā)現(xiàn)Koi42貢獻(xiàn)率較大,經(jīng)Fisher精確性檢驗(yàn),發(fā)現(xiàn)其與性別存在關(guān)聯(lián),其加性效應(yīng)接近顯著水平(P0.05)。檢測的多個上位效應(yīng)組分中,僅有D-Loop253和Koi42的加性效應(yīng)間的互作達(dá)到顯著水平。對遺傳方差組分進(jìn)行剖分發(fā)現(xiàn),加性方差占11.4%,兩個位點(diǎn)的加性效應(yīng)構(gòu)成的上位效應(yīng)占到77.5%,因此這3個分子標(biāo)記對體重的影響主要以上位效應(yīng)為主,而且是兩兩加性效應(yīng)的占比較大,可以推斷出D-Loop 253和Koi42兩個分子標(biāo)記的上位效應(yīng)起主要作用。綜上所述結(jié)果提示Koi42及與其有互作的D-Loop253可用于黃河鯉新品系的標(biāo)記輔助選擇,可以開展多個分子標(biāo)記的富集選擇。
