山羊FGF21基因克隆及其在肌內(nèi)脂肪細胞中的表達模式研究

【摘要】:本研究旨在獲得山羊FGF21基因序列,闡明其組織表達特性,同時檢測FGF21基因與FGF受體(FGFR)在山羊肌內(nèi)前體脂肪細胞分化過程中的表達水平。以簡州大耳羊為試驗動物,采用II型膠原酶消化獲得山羊原代肌內(nèi)前體脂肪細胞,采用RT-PCR技術克隆山羊FGF21基因,利用熒光定量PCR(qPCR)技術檢測FGF21基因在成年山羊不同組織中的表達特性及FGF21與FGF受體在山羊肌內(nèi)前體脂肪細胞分化過程中的表達差異。結果,克隆獲得山羊FGF21基因(GenBank登錄號:KT288195)全長序列666bp,其中開放閱讀框630bp,編碼209個氨基酸。FGF21mRNA在山羊心、肝、脾、肺、腎、脂肪和背最長肌中均檢測到表達,且在肝和脂肪中存在較高水平表達。FGF21基因與FGFR1、FGFR2表達模式相同,均是在誘導分化第2天的肌內(nèi)脂肪細胞中表達水平最高,極顯著高于其他天數(shù)(P0.01)。FGF21基因在山羊脂肪組織中存在較高水平的表達,并且在誘導分化的肌內(nèi)前體脂肪細胞成脂分化第2天表達水平最高,推測其可能通過受體FGFR1或FGFR2在山羊肌內(nèi)前體脂肪細胞分化早期發(fā)揮調(diào)控作用,此研究結果為進一步揭示FGF21基因在山羊肌內(nèi)脂肪沉積中的作用機制提供了數(shù)據(jù)支持。
[Abstract]:The aim of this study was to obtain the goat FGF21 gene sequence and to elucidate the expression characteristics of goat FGF21 gene, and to detect the expression level of FGF21 gene and FGF receptor (FGFR) during the differentiation of goat intramuscular precursor adipocytes. Goat intramuscular preadipocytes were obtained by II collagenase digestion and RT-PCR technique was used to clone goat FGF21 gene. The expression characteristics of FGF21 gene in different tissues of adult goat and the difference of expression of FGF21 and FGF receptor in the differentiation of goat intramuscular precursor adipocytes were detected by fluorescence quantitative PCR (qPCR) technique. Results the full-length sequence of goat FGF21 gene (GenBank accession number: KT288195) was 666 BP. The open reading frame 630 BP encoded 209 amino acids. The expression of FGF21mRNA was detected in heart, liver, spleen, lung, kidney, fat and longissimus dorsi muscle. The expression pattern of FGF21 gene was the same as that of FGFR1,FGFR2, and the expression level of FGF21 gene was the highest in intramuscular adipocytes on the second day of differentiation. The expression of FGF21 gene in adipose tissue of goat was higher than that in other days (P0.01), and the expression level of FGF21 gene was the highest on the second day of adipogenic differentiation of intramuscular precursor adipocytes induced by differentiation. It is speculated that it may play a regulatory role in the early differentiation of goat intramuscular precursor adipocytes by receptor FGFR1 or FGFR2. The results provide data support for further revealing the mechanism of the role of FGF21 gene in goat intramuscular fat deposition.
