轉(zhuǎn)錄組與蛋白質(zhì)組關(guān)聯(lián)分析鑒定桑樹(shù)硬枝扦插皮部生根相關(guān)功能基因

【摘要】:前期以桑品種育711號(hào)硬枝扦插皮部生根的插穗為材料,對(duì)扦插生根過(guò)程起始階段(S0)、根原基形成階段(S1)和不定根伸長(zhǎng)階段(S2)的插穗基部皮層進(jìn)行了轉(zhuǎn)錄組與蛋白質(zhì)組測(cè)序。在此基礎(chǔ)上對(duì)差異表達(dá)基因與蛋白質(zhì)(差異系數(shù)1.5,錯(cuò)誤發(fā)現(xiàn)率≤0.05)做關(guān)聯(lián)分析。結(jié)果顯示:不同階段插穗基部皮層的基因和蛋白質(zhì)表達(dá)趨勢(shì)總體上呈正相關(guān),關(guān)聯(lián)系數(shù)分別為0.470 3(S0-vs-S1)與0.113 0(S1-vs-S2);基因與蛋白質(zhì)表達(dá)趨勢(shì)相反的關(guān)聯(lián)數(shù)量較少,關(guān)聯(lián)系數(shù)分別為-0.319 9(S0-vs-S1)與-0.107 1(S1-vs-S2);基因與蛋白質(zhì)表達(dá)趨勢(shì)相同的關(guān)聯(lián)數(shù)量相對(duì)較多,關(guān)聯(lián)系數(shù)分別為0.855 4(S0-vs-S1)與0.834 6(S1-vs-S2),均呈強(qiáng)相關(guān)。在關(guān)聯(lián)分析表達(dá)趨勢(shì)相同組中篩選鑒定出21個(gè)差異表達(dá)基因,并進(jìn)行功能注釋與信號(hào)通路分析。其中,有5個(gè)基因在生根過(guò)程中持續(xù)上調(diào)表達(dá),主要包括酚氧化酶(PPO)、馬鈴薯糖蛋白(Patatin)、生長(zhǎng)素酰胺水解酶(ILL)、肌氨酸氧化酶(SOX)的編碼基因;有13個(gè)基因在根原基形成期上調(diào)表達(dá),在不定根伸長(zhǎng)期下調(diào)表達(dá),主要包括赤霉素雙加氧酶(GA3OX4)、12-氧植物二烯酸還原酶(OPR)、內(nèi)切葡聚糖酶(EG)和吲哚乙酸氨基化合成酶(GH3)的編碼基因;有3個(gè)基因在根原基形成期下調(diào)表達(dá),在不定根伸長(zhǎng)期上調(diào)表達(dá),分別為細(xì)胞膜相關(guān)鈣結(jié)合蛋白(PCa P)、肌肉LIM蛋白(MLP)和膜聯(lián)蛋白(ANX)的編碼基因。上述關(guān)聯(lián)分析的結(jié)果為深入認(rèn)識(shí)桑樹(shù)插穗皮部生根過(guò)程的生理機(jī)制提供了相關(guān)功能基因信息。
