【摘要】：木質(zhì)素是植物細(xì)胞壁的成分之一,其含量高低影響到細(xì)胞壁的彈性。橡膠樹(shù)乳管膨壓是割膠后膠乳排出的初動(dòng)力,乳管細(xì)胞壁的物理性能可能是影響乳管膨壓的因素之一。本研究采用QRT-PCR和RACE技術(shù),從巴西橡膠樹(shù)無(wú)性系CATAS7-33-97的乳管細(xì)胞中克隆到一個(gè)木質(zhì)素合成關(guān)鍵酶CCo AOMT基因的同源基因,命名為HbCCoAOMT。該基因的開(kāi)放閱讀框?yàn)?41 bp,編碼1個(gè)由246個(gè)氨基酸殘基組成的蛋白質(zhì)。該蛋白質(zhì)分子量為27.76 ku,理論等電點(diǎn)為5.255。實(shí)時(shí)熒光定量PCR結(jié)果表明,割膠和乙烯利處理均下調(diào)HbCCoAOMT基因在膠乳中的表達(dá)。該基因的表達(dá)量以及內(nèi)層樹(shù)皮的木質(zhì)素含量在兩個(gè)橡膠樹(shù)品種(CATAS8-79和PR107)間存在顯著差異。HbCCoAOMT差異表達(dá)可能影響到乳管細(xì)胞木質(zhì)素含量和乳管細(xì)胞壁的物理性能。
[Abstract]:Lignin is one of the components of plant cell wall, and its content affects the elasticity of cell wall. The pressure of rubber latex tube is the initial driving force of latex ejection after tapping, and the physical properties of the cell wall of latex tube may be one of the factors affecting the pressure of latex tube expansion. In this study, QRT-PCR and RACE techniques were used to clone a homologous gene of CCo AOMT gene, a key enzyme of lignin biosynthesis, from the latex tube cells of CATAS7-33-97 clone of Hevea brasiliensis, named HbCCoAOMT.. The open reading frame of the gene encodes a protein composed of 246 amino acid residues for 741 bp,. The molecular weight of the protein is 27.76 ku, and the theoretical isoelectric point is 5.255. The results of real-time fluorescence quantitative PCR showed that both tapping and ethephon treatment could down-regulate the expression of HbCCoAOMT gene in latex. There were significant differences in the expression of the gene and the lignin content in the inner bark between the two Hevea varieties (CATAS8-79 and PR107). The differential expression of HbCCoAOMT might affect the lignin content and the physical properties of the cell wall of milk tube.
