齊口裂腹魚白介素-1β基因的克隆及表達譜

【摘要】:探討齊口裂腹魚白介素-1β(Interleukin-1β,IL-1β)基因的基本特點。用RT-PCR方法以齊口裂腹魚脾臟c DNA作為模板擴增IL-1β基因,用生物信息學軟件分析其序列,用RT-PCR方法檢測IL-1β基因在齊口裂腹魚6種組織的表達情況。克隆獲得的IL-1β序列長1 252 bp,共編碼276個氨基酸。Gen Bank序列號為KU886235。IL-1β蛋白相對分子量為31.25 k D,等電點5.45,預測為親水性蛋白。二級結構主要結構元件為隨機卷曲和延伸鏈,有4個N-糖基化位點和16個磷酸化位點。NJ法系統進化樹顯示,齊口裂腹魚與鯉魚、鯽魚的親緣關系最近。組織表達分析顯示,IL-1β基因主要在脾臟和腎臟中表達。這為深入研究魚IL-1β的生物學功能提供理論依據。
[Abstract]:To study the basic characteristics of interleukin-1 尾 (Interleukin-1 尾) gene in fish. The IL-1 尾 gene was amplified by using c DNA of the spleen of the fish by RT-PCR method. The sequence of IL-1 尾 gene was analyzed by bioinformatics software. The expression of IL-1 尾 gene in 6 kinds of tissues was detected by RT-PCR method. The cloned IL-1 尾 sequence (1 252 bp,) encodes 276 amino acids. Gen Bank sequence number is 31.25 KD, isoelectric point 5.45, predicted to be hydrophilic protein. The main structural elements of the secondary structure were random curl and extension chain. There were 4 N-glycosylation sites and 16 phosphorylation sites. NJ phylogenetic tree showed that the phylogenetic relationship between Carp and Carp, Carassius auratus was the closest. Tissue expression analysis showed that IL-1 尾 gene was mainly expressed in spleen and kidney. This provides a theoretical basis for the further study of the biological function of fish IL-1 尾.
