水稻抗稻瘟病基因Pita特異性分子標(biāo)記開發(fā)及應(yīng)用

【摘要】:Pita是一個對稻瘟病生理小種具有廣譜抗性的基因,通過對Pita抗病等位基因與感病等位基因編碼區(qū)進(jìn)行PCR擴(kuò)增、測序與序列比對,在編碼區(qū)的+2 001 bp即內(nèi)含子中鑒定到一處特異性編碼序列(抗、感病等位基因分別為GCC和CTAT),針對該目標(biāo)位點(diǎn),本研究利用兩對交叉引物PCR(PCR with confronting two-pair primers,PCR-CTPP)方法開發(fā)出一套鑒定該目標(biāo)位點(diǎn)的共顯性標(biāo)記用于鑒定Pita等位基因類型,結(jié)果表明該標(biāo)記特異性強(qiáng)、穩(wěn)定、結(jié)果準(zhǔn)確、耗時(shí)少、成本低廉,可廣泛應(yīng)用于分子標(biāo)記輔助選擇育種或水稻資源中對Pita基因型鑒定、篩選的應(yīng)用。
[Abstract]:Pita is a broad-spectrum resistance gene to rice blast physiological races. The coding region of Pita resistance allele and susceptible allele was amplified by PCR, sequenced and sequenced. A specific coding sequence was identified in the 2 001 BP intron of the coding region. The anti- and susceptible alleles were GCC and CTAT), for the target site, respectively. In this study, a set of co-dominant markers to identify the target locus was developed by using two pairs of cross-primer PCR (PCR with confronting two-pair primer PCR-CTPP to identify the allele type of Pita. The results showed that the marker was specific, stable, accurate, time-consuming and low cost. It can be widely used in molecular marker-assisted selection breeding or in the identification and screening of Pita genotypes in rice resources.
