不同鋅源及添加水平對(duì)斷奶仔豬微量元素代謝與鋅相關(guān)基因表達(dá)的影響

【摘要】:本文旨在研究不同水平的蛋氨酸鋅(Zn-Met)與一水硫酸鋅(ZnSO4·H2O)組合使用對(duì)斷奶仔豬生產(chǎn)性能、血液指標(biāo)、免疫指標(biāo)、微量元素代謝及鋅相關(guān)基因表達(dá)量的影響,以確定有機(jī)鋅與無(wú)機(jī)鋅的最佳配合模型,為養(yǎng)殖生產(chǎn)中低水平添加鋅滿足斷奶仔豬的生長(zhǎng)需要,減少糞便鋅的排放提供理論依據(jù)。選擇30日齡、平均體重(10.69±0.26)kg、健康狀態(tài)良好的三元公豬35頭,隨機(jī)分為5個(gè)處理,每個(gè)處理7個(gè)重復(fù),每個(gè)重復(fù)1頭豬。采用基礎(chǔ)飼糧相同(預(yù)混料不添加鋅)。處理Ⅰ為對(duì)照組(基礎(chǔ)飼糧+硫酸鋅100 mg/kg),處理Ⅱ-Ⅴ為試驗(yàn)組,分別用蛋氨酸鋅以1:2的比例替代對(duì)照組中的硫酸鋅25 mg/kg、50 mg/kg、75 mg/kg與100 mg/kg(添加水平均以鋅元素計(jì))。試驗(yàn)期共49 d,預(yù)試期7 d,正試期42 d。試驗(yàn)結(jié)果表明:(1)與對(duì)照組相比,試驗(yàn)組平均日增重、平均日采食、料重比差異均不顯著(P0.05)。(2)與對(duì)照組相比,試驗(yàn)組的鋅表觀消化率顯著提高(P0.05),有機(jī)物消化率無(wú)顯著差異(P0.05)。(3)與單一的蛋氨酸鋅或者硫酸鋅相比,混合鋅能提高仔豬血清白細(xì)胞(WBC)含量與淋巴細(xì)胞(LYM)含量,且其中添加50 mg/kg硫酸鋅組合25 mg/kg蛋氨酸鋅的試驗(yàn)組相比其他組有著顯著提高(P0.05)。(4)與對(duì)照組相比,添加混合鋅的試驗(yàn)組血清鋅含量、血清錳含量、肝臟鋅水平與胰臟鋅水平均顯著升高(P0.05),糞便鋅的排泄量顯著降低(P0.05),說(shuō)明混合鋅能改善仔豬血清、組織中的微量元素狀況,顯著降低糞便鋅的排泄量。(5)對(duì)照組的胸腺、脾臟與胰臟的器官指數(shù)最低,與對(duì)照組相比,試驗(yàn)組的胸腺、脾臟、胰臟指數(shù)均顯著升高(P0.05),血清MDA顯著降低(P0.05),說(shuō)明混合鋅能提高仔豬抗應(yīng)激能力。(6)與對(duì)照組相比,添加混合鋅的試驗(yàn)組的各組織金屬硫蛋白(MT)mRNA與脾臟、腎臟的C-反應(yīng)蛋白(CRP)mRNA相對(duì)表達(dá)量顯著升高(P0.05);脾臟與腎臟的結(jié)合珠蛋白(HP)mRNA與α1-酸性糖蛋白(AGP)mRNA、各組織血清淀粉樣蛋白A(SAA)mRNA表達(dá)量均顯著高于對(duì)照組,且均隨蛋氨酸鋅添加量升高而升高(P0.05),說(shuō)明蛋氨酸鋅與硫酸鋅混合使用能提高各組織MT與CRP的合成與分泌,蛋氨酸鋅更能誘導(dǎo)急性期蛋白(APP)基因表達(dá),提高免疫力。綜上所述,蛋氨酸鋅與硫酸鋅混合使用可以顯著改善斷奶仔豬機(jī)體鋅營(yíng)養(yǎng)水平。與單一使用硫酸鋅相比,蛋氨酸鋅與硫酸鋅混合使用不僅能在減少飼料添加鋅水平,還能提高機(jī)體抗病力,改善仔豬血清、組織中的微量元素狀況,顯著降低糞便鋅的排泄量,其中以50 mg/kg的硫酸鋅組合25 mg/kg的蛋氨酸鋅的微量元素減排效果最佳;蛋氨酸鋅與硫酸鋅混合使用可以增強(qiáng)MT分泌,蛋氨酸鋅比雙倍水平硫酸鋅更能誘導(dǎo)APP mRNA表達(dá)量,增強(qiáng)斷奶仔豬非特異性免疫機(jī)制。
