【摘要】：以207株‘金冠’與‘富士’蘋果的F1雜交分離群體實(shí)生樹為試材,挑選抗炭疽菌葉枯病基因R_(gls)位點(diǎn)區(qū)域內(nèi)的,第15條染色體上,位于SSR標(biāo)記S0304673和S0405127之間500 kb物理距離內(nèi),由全基因組重測序技術(shù)所獲得的與抗該病相關(guān)的10個SNP及10個InDel標(biāo)記,通過高分辨率熔解曲線(High Resolution Melting,HRM)技術(shù)篩選出與基因R_(gls)位點(diǎn)緊密連鎖的2個SNP及4個InDel標(biāo)記。通過對重組個體的基因型及表型分析,發(fā)現(xiàn)標(biāo)記InDel4227和InDel4254表現(xiàn)出與R_(gls)基因位點(diǎn)共分離,將基因R_(gls)精細(xì)定位于標(biāo)記InDel4199和SNP4299之間100 kb的物理距離內(nèi)。對該基因位點(diǎn)兩側(cè)4.1~4.3Mb距離內(nèi)的基因進(jìn)行統(tǒng)計分析,表明在該區(qū)段內(nèi)存在40個有功能注釋的基因,涉及到細(xì)胞組成、分子功能和生物過程方面共19個GO分類。
[Abstract]:In the F _ 1 hybrid population of 'Jinguan' and 'Fuji' apple, selected the region of resistance to anthrax leaf blight gene R _ (gls), on chromosome 15, located within 500kb physical distance between SSR marker S0304673 and S0405127. Ten SNP and 10 InDel markers associated with the disease resistance were obtained from the whole genome resequencing technique. Two SNP and four InDel markers closely linked to the gene R _ (gls) locus were screened by high-resolution fusion curve (High Resolution melting HRM. By genotypic and phenotypic analysis of the recombinant individuals, it was found that the marker InDel4227 and InDel4254 were coisolated from the R _ (gls) gene locus, and the gene R _ (gls) was located within the physical distance of 100kb between the labeled InDel4199 and SNP4299. The statistical analysis of the genes in the 4.1~4.3Mb distance between the two sides of the gene site showed that there were 40 functional annotated genes in this region, involving 19 go classifications in cell composition, molecular function and biological processes.
