【摘要】：目的比較電針、手針長強(qiáng)穴對FMR1基因敲除小鼠皮質(zhì)區(qū)環(huán)磷腺苷效應(yīng)元件結(jié)合蛋白(CREB)mRNA及腦源性神經(jīng)營養(yǎng)因子(BDNF)mRNA表達(dá)量的影響,并探討其作用機(jī)制。方法選取適齡FMR1基因敲除型小鼠18只,隨機(jī)分為空白組、手針組、電針組,每組6只。取穴長強(qiáng)穴,手針組在留針期間每隔10min行手法運(yùn)針15s,電針組在手針針刺基礎(chǔ)上連接電針儀,施以連續(xù)波,頻率2Hz,留針30min,每日1次,連續(xù)治療14 d;空白組在同等條件下飼養(yǎng),并僅模擬抓取,不予任何干預(yù)。行為學(xué)實(shí)驗(yàn)觀察小鼠空間學(xué)習(xí)記憶能力及其自主活動(dòng)次數(shù),熒光定量PCR檢測小鼠皮質(zhì)區(qū)CREB mRNA及BDNF mRNA表達(dá)量。結(jié)果水迷宮實(shí)驗(yàn)中針刺治療前后小鼠逃避潛伏期具有顯著差異(P0.05),且針刺干預(yù)后,手針組逃避潛伏期比電針組明顯縮短(P0.05),手針組原平臺象限時(shí)間/逃避潛伏期比電針組明顯延長(P0.05)。與電針組比較,手針組皮質(zhì)區(qū)BDNF mRNA表達(dá)量明顯增加(P0.05);與空白組對比,電針組和手針組皮質(zhì)區(qū)CREB mRNA表達(dá)量無明顯差異(P0.05);結(jié)論針刺"長強(qiáng)"穴,手針組能顯著影響FMR1基因敲除小鼠的學(xué)習(xí)記憶能力及異常行為活動(dòng),其機(jī)制可能與調(diào)控中樞神經(jīng)的BDNF mRNA、CREB mRNA表達(dá)量有關(guān)。
[Abstract]:Objective to compare the effects of electroacupuncture and hand acupuncture on the expression of cyclophosphate adenosine effector element binding protein (CREB) mRNA and brain-derived neurotrophic factor (BDNF) mRNA in the cortex of FMR1 gene knockout mice. Methods Eighteen FMR1 knockout mice of appropriate age were randomly divided into blank group, hand acupuncture group and electroacupuncture group with 6 mice in each group. The long and strong points were taken from the acupoints, the hand acupuncture group operated the acupuncture at intervals of 10min every other time for 15 s, the electroacupuncture group was connected with the electroacupuncture instrument on the basis of the hand acupuncture, and the continuous wave was applied at the frequency of 2 Hz, the needle was kept for 30 minutes, once a day for 14 days, and the blank group was fed under the same conditions. And only simulated grab, no intervention. Behavioral experiments were performed to observe the spatial learning and memory ability and the number of autonomic activities in mice. The expressions of CREB mRNA and BDNF mRNA in the cortex of mice were detected by fluorescence quantitative PCR. Results in the water maze experiment, there was significant difference in the escape latency of mice before and after acupuncture treatment (P0.05). After acupuncture intervention, the escape latency of the hand acupuncture group was significantly shorter than that of the electroacupuncture group (P0.05), and the original platform quadrant time / escape latency of the hand acupuncture group was significantly longer than that of the electroacupuncture group (P0.05). Compared with the electroacupuncture group, the expression of BDNF mRNA in the cortex of the hand acupuncture group was significantly increased (P0.05), but there was no significant difference in the expression of CREB mRNA between the electroacupuncture group and the hand acupuncture group (P0.05). The ability of learning and memory and abnormal behavior of FMR1 knockout mice were significantly affected by hand acupuncture group. The mechanism may be related to the regulation of the expression of BDNF mRNA-CREB mRNA in the central nervous system.
