【摘要】：本研究采用RT-PCR技術克隆了藥材甲熱激蛋白60(Heat shock protein 60,Hsp60)基因的cDNA全長序列,命名為SpHsp60(GenBank登錄號:KX778623).氨基酸序列分析表明SpHsp60具有Hsp60基因家族高度保守的基序,實時定量PCR技術檢測了不同溫度脅迫后該基因的表達量,結(jié)果表明:-5℃和0℃低溫處理2h,藥材甲成蟲體內(nèi)的SpHsp60的表達量均顯著高于對照組,且42℃高溫處理2h后的表達量也顯著高于對照組.說明SpHSP60與藥材甲應對極端溫度脅迫相關.
[Abstract]:In this study, the full-length cDNA sequence of heat shock protein 60 Hsp60 gene was cloned by RT-PCR and named SpHsp60 (GenBank accession number: KX778623). Amino acid sequence analysis showed that SpHsp60 had a highly conserved motif of Hsp60 gene family. The expression of SpHsp60 gene was detected by real-time quantitative PCR under different temperature stress. The results showed that the expression of SpHsp60 was significantly higher than that of the control group at the temperature of 5 鈩，
