鄱陽(yáng)湖泥鰍黑色素聚集激素1基因及其蛋白結(jié)構(gòu)特征分析

【摘要】:為研究黑色素聚集激素(Melanin-concentrating hormone,MCH)在泥鰍(Misgurnus anguillicaudatus)皮膚黑色素沉著過程中所起的作用,研究采用RACE-PCR技術(shù)首次克隆出了鄱陽(yáng)湖泥鰍前原黑色素聚集激素1(Prepro-melanin-concentrating hormone 1,Pmch1)基因c DNA全長(zhǎng)序列,將其命名為Ma Pmch1。運(yùn)用生物信息學(xué)軟件對(duì)Ma Pmch1基因及其蛋白的理化性質(zhì)和結(jié)構(gòu)特征進(jìn)行了分析,構(gòu)建了系統(tǒng)進(jìn)化樹。其c DNA全長(zhǎng)為570 bp,存在一個(gè)長(zhǎng)度為110 bp的Cp G島,位于263—372 bp;該基因開放閱讀框共375 bp,編碼含124個(gè)氨基酸的蛋白質(zhì),共有7個(gè)潛在的磷酸化位點(diǎn);PMCH1蛋白經(jīng)水解產(chǎn)生17個(gè)氨基酸的環(huán)形神經(jīng)肽黑色素聚集激素1(MCH1)。PMCH1和MCH1均為親水性蛋白,二級(jí)結(jié)構(gòu)以無規(guī)則卷曲為主;MCH1超二級(jí)結(jié)構(gòu)具有一個(gè)反平行β-折疊片層,通過二硫鍵連接形成環(huán)形神經(jīng)肽。同源性分析表明鄱陽(yáng)湖泥鰍MCH1(Ma MCH1)氨基酸序列與其他硬骨魚類的高度一致。系統(tǒng)進(jìn)化樹顯示,包括泥鰍在內(nèi)的鯉形目PMCH1聚為一支,且物種間PMCH1的親緣關(guān)系與傳統(tǒng)的分類地位相吻合。在脊椎動(dòng)物中,MCH存在高度保守的序列RCM*GRVYRPCW(*代表隨機(jī)氨基酸),證明該基因在進(jìn)化上是極為保守的。
[Abstract]:In order to study the role of Melanin-concenting hormone (MCH) in the process of melanosis in the skin of Misgurnus anguillicaudatus, the full-length cDNA sequence of premelanin aggregation hormone 1Pmch1 gene of Poyang Lake Loach was cloned by RACE-PCR for the first time. It was named Ma Pmch1. The physicochemical properties and structural characteristics of Ma Pmch1 gene and its protein were analyzed by bioinformatics software, and a phylogenetic tree was constructed. The total length of c DNA is 570 BP, and there is a 110 BP CP G island located at 263-372 BP. The open reading frame of this gene is 375 BP and encodes a protein containing 124 amino acids. A total of 7 potential phosphorylation sites of PMCH1 protein were hydrolyzed to produce a 17 amino acid ring neuropeptide melanin aggregation hormone 1 (MCH1). PMCH1 and MCH1 were hydrophilic proteins. The secondary structure is mainly composed of irregular curls and MCH1 has an anti-parallel 尾 -folded lamellae, which is connected by disulfide bond to form a ring neuropeptide. Homology analysis showed that the amino acid sequence of MCH1 (Ma MCH1) of Poyang Lake Loach was highly consistent with that of other bony fishes. Phylogenetic tree showed that the Cyprinidae PMCH1, including Loach, were clustered into one branch, and the phylogenetic relationship of PMCH1 between species was consistent with the traditional taxonomic position. The existence of highly conserved sequence RCM-GRVYRPCW (* representing random amino acids) in vertebrates suggests that the gene is extremely conserved in evolution.
