【摘要】：目的:研究Bmi-1基因?qū)κ彻荀[癌細胞增殖、侵襲、耐藥的作用,并探討其可能機制。方法:利用Bmi-1 siRNA轉(zhuǎn)染食管癌細胞Eca9706,采用Westen-blot、CCK-8檢測細胞增殖能力的改變,Transwell小室檢測細胞侵襲能力的改變,并檢測順鉑耐藥性的改變。結(jié)果:Bmi-1 siRNA轉(zhuǎn)染食管癌細胞Eca9706后,細胞增殖能力、侵襲能力均顯著下降(P0.05),且順鉑耐藥性顯著下降。結(jié)論:Bmi-1基因具有促進食管癌細胞增殖、侵襲的作用,并促進順鉑耐藥的作用。
[Abstract]:Aim: to study the effect of Bmi-1 gene on proliferation, invasion and drug resistance of esophageal squamous cell carcinoma cells. Methods: esophageal carcinoma cells Eca9706 were transfected with Bmi-1 siRNA. The changes of cell proliferation and cisplatin resistance were detected by Westen-blottCCK-8. Results the proliferation and invasion ability of human esophageal carcinoma cell line Eca9706 transfected with 10% Bmi-1 siRNA decreased significantly (P 0.05), and cisplatin resistance decreased significantly. Conclusion: Bmi-1 gene can promote the proliferation and invasion of esophageal cancer cells and promote cisplatin resistance.
