【摘要】：作為植物信號分子的茉莉酸(JA)參與植物的發(fā)育與代謝過程,丙二烯環(huán)化酶(AOC)是JA合成途徑的關鍵酶。在本研究中,利用同源克隆結合RACE技術從金魚草中克隆AOC基因,命名為Am AOC。Am AOC基因開放閱讀框長762 bp,由254個氨基酸殘基組成,該基因包含一個Alleneoxcyc結構域,有兩個內(nèi)含子和三個外顯子。同源比對分析結果表明Am AOC與其他植物的AOC有較高的同源性。Am AOC基因在不同組織中的表達分析發(fā)現(xiàn)金魚草花中的Am AOC相對表達量較高,不同花期金魚草RT-PCR表明,花蕾期的Am AOC相對表達量最高,盛花期的不同花器官熒光定量PCR分析表明,金魚草花筒和花萼中Am AOC相對表達量較高。
[Abstract]:JAA, a plant signaling molecule, is involved in the development and metabolism of plants. Allyl cyclase (AOC) is the key enzyme in JA synthesis pathway. In this study, the AOC gene was cloned by homologous cloning and RACE from Goldfish. The gene was named am AOC.Am AOC open reading frame 762 BP and consisted of 254 amino acid residues. The gene contains a Alleneoxcyc domain. There are two introns and three exons. The results of homology analysis showed that am AOC had high homology with other plants' AOC. The expression of am AOC gene in different tissues was higher than that in other plants. The relative expression of am AOC in flower bud stage was the highest, and the fluorescence quantitative PCR analysis of different floral organs at full flowering stage showed that the relative expression of am AOC was higher in the tube and calyx of Astragalus mandshurica.
