水母雪蓮表皮毛發(fā)育相關基因SmTTG1的遺傳轉化研究

【摘要】:水母雪蓮(Saussurea medusa Maxim),又稱水母雪兔子,屬菊科風毛菊屬,是一種生長于青藏高原地區(qū)的珍稀、瀕危藥用植物。同時,它又是一種典型的高山植物。植株密被表皮毛是水母雪蓮最具有代表性的形態(tài)特征,它是水母雪蓮對極端環(huán)境的一種適應結構。研究水母雪蓮表皮毛的形成和發(fā)育過程,對揭示高山植物適應環(huán)境的特殊機制將具有重要的理論意義。為此,本研究以水母雪蓮作為實驗材料,以表皮毛發(fā)育相關基因SmTTG1為研究對象,構建了SmTTG1基因的植物表達載體,通過根癌農桿菌為介導,轉入普通煙草中。主要研究結果如下:(1)根據(jù)本實驗室已克隆得到的水母雪蓮SmTTG1基因序列(Genbank登陸號:KX447632),在其CDS兩側分別設計上下游引物,在上游引物5′端加入酶切位點SalⅠ,在下游引物5′端加入酶切位點Bam HⅠ,克隆了水母雪蓮SmTTG1基因的CDS序列(含酶切位點)。(2)通過酶切法,將目的基因SmTTG1插入質粒pRI 101-AN DNA的多克隆位點,構建了植物表達載體pRI 101-AN-SmTTG1,并通過凍融法將其導入農桿菌C58C1中。(3)在根癌農桿菌介導下,通過葉盤法將目的基因SmTTG1轉入普通煙草中。通過PCR和RT-PCR檢測,獲得8株轉基因煙草植株。形態(tài)學比較表明,轉基因煙草植株與對照植株無明顯差異,說明SmTTG1基因過量表達不一定對煙草的形態(tài)產生影響。
[Abstract]:Saussurea medusa Maxima, also called jellyfish snow rabbit, is a rare and endangered medicinal plant in the Qinghai-Tibet Plateau. At the same time, it is a typical alpine plant. The dense coat of the plant is the most representative morphological feature of the jellyfish Saussurea, which is an adaptive structure of the jellyfish to the extreme environment. It is of great theoretical significance to study the formation and development of the epidermal hair of Saussurea jellyfish in order to reveal the special mechanism of adaptation of alpine plants to the environment. Therefore, the plant expression vector of SmTTG1 gene was constructed by using Saussurea jellyfish as experimental material and epidermal development related gene SmTTG1 as the research object. The vector was transferred into common tobacco by Agrobacterium tumefaciens mediated by Agrobacterium tumefaciens.
