【摘要】：目的探討新疆地區(qū)BRAF V600E基因點(diǎn)突變在甲狀腺乳頭狀癌(papillary thyroid carcinoma,PTC)中的表達(dá),及其與臨床病理特征的關(guān)系。方法收集原發(fā)性PTC 178例,石蠟包埋組織經(jīng)QIAGEN試劑盒提取DNA,經(jīng)Taq Man探針熒光定量PCR技術(shù)檢測PTC中BRAF基因點(diǎn)突變。結(jié)果 178例原發(fā)性PTC中BRAF V600E基因點(diǎn)突變142例,突變率為79.8%(142/178)�；颊吣挲g(≥45歲)、腫瘤直徑(≤1 cm)、被膜外侵犯BRAF V600E基因點(diǎn)突變率高(P均0.05),而BRAF V600E基因點(diǎn)突變與患者性別、多灶性及淋巴結(jié)轉(zhuǎn)移灶和腫瘤部位均無相關(guān)性(P均0.05)。結(jié)論新疆地區(qū)PTC中BRAF V600E基因點(diǎn)突變與臨床病理特征密切相關(guān),且具有一定特異性,可作為PTC診斷提供依據(jù)。
[Abstract]:Objective to investigate the expression of BRAF V600E gene point mutation in papillary thyroid carcinoma of thyroid and its relationship with clinicopathological features. Methods A total of 178 cases of primary PTC were collected. DNA was extracted from paraffin embedded tissue by QIAGEN kit. The point mutation of BRAF gene in PTC was detected by Taq Man probe fluorescence quantitative PCR technique.
