【摘要】：甘蔗(Saccharum officinarum)是制造蔗糖、提煉乙醇的主要原料。隨著甘蔗轉(zhuǎn)基因育種的開展,建立一套快速精準(zhǔn)轉(zhuǎn)基因分子檢測技術(shù)在甘蔗轉(zhuǎn)基因研究中有重要的應(yīng)用前景。本研究以二價(jià)抗蟲轉(zhuǎn)基因甘蔗為材料,利用多重PCR建立同時(shí)檢測Bar基因、Bt基因和馬鈴薯蛋白酶抑制劑基因(PinⅡ基因)。研究表明,該多重PCR檢測體系的最佳退火溫度為53℃,有效擴(kuò)增Bar基因(415 bp)、Bt基因(780 bp)和PinⅡ(1 000 bp)3個(gè)基因片段。多重PCR技術(shù)進(jìn)行外源基因的檢測,節(jié)省時(shí)間、降低成本、提高效率并可以對(duì)該轉(zhuǎn)化植物進(jìn)行特異性鑒定。
