篩選試劑紅細胞的血型基因分型方法建立和應用

【摘要】:目的建立重要的紅細胞血型的基因分型技術用于篩選抗原譜合理的試劑紅細胞。方法首先建立檢測紅細胞血型系統(tǒng)Rh CE、Kidd、MNS、Duffy、Diego、Lutheran、Colton、Lutheran和Kell等的聚合酶鏈式反應-序列特異性引物(PCR-SSP)分型技術;對于無法獲得相關血型試劑的Diego和Colton血型系統(tǒng),采用測序方法驗證該技術檢測血型的準確性,其他6個血型系統(tǒng)則用血清學方法驗證該技術檢測血型的準確性;最后應用該方法對82名O型固定獻血者的8個血型系統(tǒng)做基因分型。結果血型基因PCR-SSP分型技術的結果,與血清學和測序檢測結果比較,完全一致。所建立的血型基因分型技術對82名O型獻血者8種紅細胞血型檢測,篩選出13位可能適合提供試劑紅細胞的獻血者。結論成功建立了分型準確、方便、經(jīng)濟的紅細胞血型基因PCR-SSP技術,用于從大量獻血者中篩選出抗原譜合理的試劑紅細胞。
[Abstract]:Objective to establish an important genotyping technique of erythrocyte blood group for screening reagent erythrocytes with reasonable antigen spectrum.Methods the polymerase chain reaction-sequence specific primer (PCR-SSPP) typing technique was established for the detection of erythrocyte blood group system Rh CED MNS Duffy Diegol Lutheran and Kell, and the Diego and Colton blood group systems which could not obtain the relevant blood group reagents were used.The accuracy of this technique was verified by sequencing method, and the other 6 blood group systems were tested by serological method.Finally, 8 blood group systems of 82 O-type fixed blood donors were genotyped by this method.Results the results of PCR-SSP typing were consistent with those of serology and sequencing.The established genotyping technique was used to detect 8 kinds of erythrocyte blood groups of 82 O blood donors and 13 blood donors who might be suitable for providing reagent red blood cells were selected.Conclusion the accurate, convenient and economical PCR-SSP technique of erythrocyte blood group gene has been successfully established, which can be used to screen the red blood cells with reasonable antigen profile from a large number of blood donors.
