流體剪切力對人脂肪基質(zhì)細(xì)胞成骨相關(guān)基因表達(dá)影響的研究

【摘要】:目的探討流體剪切力(FSS)對人脂肪基質(zhì)細(xì)胞(ADSC)成骨相關(guān)基因骨鈣素(OCN)、轉(zhuǎn)錄因子Osterix(Osx)及核心結(jié)合因子(Cbf)a1 / Runt相關(guān)轉(zhuǎn)錄因子(Runx)2表達(dá)的影響。方法將脂肪抽吸術(shù)獲取的人體脂肪進(jìn)行常規(guī)培養(yǎng)(A1、A2組)和誘導(dǎo)培養(yǎng)(A3、A4組),然后分別對其加載強(qiáng)度為0.3 Pa的FSS(A1、A3組),未加FSS的為對照組(A2、A4組),通過逆轉(zhuǎn)錄-聚合酶鏈反應(yīng)檢測骨鈣素、轉(zhuǎn)錄因子Osx及Cbfa1/Runx2基因表達(dá)的情況。結(jié)果 A3、A4組在FSS加載后,其成骨相關(guān)基因的表達(dá)增強(qiáng);而A1、A2組未見成骨特異性基因條帶。這表明在常規(guī)培養(yǎng)條件下,無論是否有FSS的刺激,均無成骨特異性基因表達(dá)。結(jié)論脂肪基質(zhì)細(xì)胞在誘導(dǎo)培養(yǎng)條件下,FSS可促使其向成骨細(xì)胞分化,可作為骨組織工程的種子細(xì)胞。
[Abstract]:Objective to investigate the effects of fluid shear stress (FSS) on the expression of osteoblast-associated gene osteocalcin (OCN), transcriptional factor Osterix Osx (Osx) and the core binding factor CbfA-1 / Runt related transcription factor (Runxt2) in human adipose stromal cells (ADSCs).Methods Human fat obtained by liposuction was cultured in routine culture group A _ (1) A _ (2) and induced culture group A _ (3) A _ (4), and then loaded with FSSA _ (1) A _ (1) A _ (3) group with a strength of 0.3 Pa and control group (group A _ (2) A _ (4) with no FSS added) by reverse transcription-polymerase chain reaction (RT-PCR).Reaction detection of osteocalcin,Expression of Osx and Cbfa1/Runx2 genes.Results the expression of osteoblast-associated genes was increased in group A _ 3 and A _ (3) A _ (4) after FSS loading, while no band of osteoblast-specific genes was found in group A _ (1) A _ (2).This indicated that there was no osteoblast-specific gene expression under conventional culture conditions, whether or not stimulated by FSS.Conclusion FSS can induce adipose stromal cells to differentiate into osteoblasts and can be used as seed cells for bone tissue engineering.
