珠江上游地區(qū)蚊蟲乙型腦炎病毒核酸檢測及PrM和E基因序列分析

【摘要】:試驗旨在了解珠江上游地區(qū)乙型腦炎病毒(JEV)主要傳播媒介蚊蟲的種類、分布及攜帶JEV基因型和分子特征。2013年7月在珠江上游地區(qū)師宗縣采集蚊蟲和庫蠓標(biāo)本,并對蚊蟲進(jìn)行分類鑒定;同時采用黃病毒屬引物對采集到的蚊蟲和庫蠓標(biāo)本進(jìn)行檢測,陽性標(biāo)本用JEV PrM和E基因特異引物進(jìn)行擴增、測序,然后采用生物信息學(xué)軟件進(jìn)行序列分析。在4個采集點共采集蚊蟲3 705只,分別屬于庫蚊、按蚊、伊蚊和阿蚊4個屬的7種蚊蟲,牛圈以中華按蚊、騷擾阿蚊為優(yōu)勢蚊種,羊圈以中華按蚊、騷擾阿蚊為優(yōu)勢蚊種,豬圈以三帶喙庫蚊、中華按蚊和騷擾阿蚊為優(yōu)勢蚊種。采用JEV PrM和E基因特異引物對采集到蚊蟲和庫蠓標(biāo)本分162批進(jìn)行擴增、測序,采用生物信息學(xué)軟件進(jìn)行序列分析。結(jié)果顯示,4株蚊蟲攜帶的病毒均為基因Ⅰ型JEV,與疫苗株SA14-14-2E蛋白存在15個氨基酸差異位點,在8個影響病毒毒力的重要位點上未發(fā)生突變。以上結(jié)果提示珠江上游地區(qū)存在多種蚊蟲種類,三帶喙庫蚊和中華按蚊是該地區(qū)的優(yōu)勢蚊種,三帶喙庫蚊是當(dāng)?shù)豃EV的主要傳播媒介,在當(dāng)?shù)氐拿浇橹写嬖诨颌裥蚃EV流行,而流行株的毒力并未降低。
[Abstract]:The aim of the experiment was to understand the species, distribution and JEV genotypes and molecular characteristics of Japanese encephalitis virus (JEV) in the upper reaches of the Pearl River. The specimens of mosquitoes and Culicomidges were collected in Shizong County, the upper reaches of the Pearl River in July 2013.At the same time, the samples of mosquitoes and Culicoides were detected by using yellow virus primer, and the positive samples were amplified and sequenced by JEV PrM and E gene specific primers.Then the sequence analysis was carried out with bioinformatics software.A total of 3 705 mosquitoes belonging to 4 genera of Culex pipiens, Anopheles aegypti and Aedes albopictus were collected in 4 collection sites. Anopheles sinensis was the dominant mosquito species in cattle circles, Anopheles sinensis as dominant species, Anopheles sinensis as dominant species and Anopheles albopictus as dominant species.Culex tritaeniorhynchus, Anopheles sinensis and Anopheles albopictus are the dominant species in pigsty.JEV PrM and E gene specific primers were used to amplify the samples of mosquitoes and Culicomidges in 162 batches, sequenced, and sequenced by bioinformatics software.The results showed that the viruses carried by the four strains were all gene type I, and there were 15 amino acid difference sites between them and the SA14-14-2E protein of the vaccine strain, and no mutation was found at the 8 important sites that affected the virulence of the virus.These results suggest that there are many mosquito species in the upper reaches of the Pearl River. Culex tritaeniorhynchus and Anopheles sinensis are the dominant species in this area. Culex tritaeniorhynchus is the main transmission vector of JEV in the region, and the gene type I JEV is prevalent in the local vectors.But the virulence of the epidemic strain did not decrease.
