牡丹開(kāi)花調(diào)控轉(zhuǎn)錄因子基因PsFUL1的克隆與表達(dá)分析

【摘要】:以‘洛陽(yáng)紅’牡丹為試驗(yàn)材料,采用RT-PCR方法從其花芽中克隆得到1個(gè)開(kāi)花調(diào)控重要轉(zhuǎn)錄因子基因FRUITFULL(FUL1)的同源基因,其c DNA開(kāi)放閱讀框長(zhǎng)度為768 bp,編碼255個(gè)氨基酸。序列比對(duì)和結(jié)構(gòu)域分析表明,此蛋白具有MADS超家族、K-box超家族的結(jié)構(gòu)域和ARG80多結(jié)構(gòu)域,且具有MADS-box和K-box的穩(wěn)定保守基序。NCBI blast分析發(fā)現(xiàn)克隆得到的氨基酸序列與葡萄中的FUL類蛋白具有較高同源性,為69%。因此將其命名為Ps FUL1,Gen Bank登錄號(hào)為KX621277。系統(tǒng)進(jìn)化樹(shù)分析表明,Ps FUL1與葡萄編碼的Vv FUL1親緣關(guān)系最近,屬于MADS基因家族中的AP1/FUL亞家族。q RT-PCR表明,Ps FUL1基因在‘洛陽(yáng)紅’牡丹不同組織中表達(dá)差異顯著,在花芽和花瓣中的表達(dá)量最高,苞片和葉片次之,根中最少。在不同品種牡丹(早花‘迎日紅’、中花‘洛陽(yáng)紅’、晚花‘花王’)的6個(gè)不同開(kāi)花時(shí)期的花瓣中,Ps FUL1基因表達(dá)量因花期早晚與開(kāi)花時(shí)期不同而差異顯著,表明Ps FUL1基因?qū)δ档ら_(kāi)花及花期的早晚有重要調(diào)控作用。
[Abstract]:A homologous gene of FRUITFULL FUL1, an important transcription factor gene, was cloned from the flower bud of 'Luoyang Red' peony by RT-PCR method. Its c DNA open reading frame length was 768 BP, encoding 255 amino acids.Sequence alignment and domain analysis showed that the protein had the domain of MADS superfamily K-box superfamily and ARG80 multi-domain.The stable conserved motif of MADS-box and K-box. NCBI blast analysis showed that the cloned amino acid sequence had high homology with the FUL class protein in grape.Therefore, it is named as PS FUL1G Bank login number is KX621277.Phylogenetic tree analysis showed that the phylogenetic relationship between PPS FUL1 and VV FUL1 encoded by grape was the most recent. AP1/FUL subfamily. Q RT-PCR, belonging to MADS gene family, showed significant difference in expression of PPS FUL1 gene in different tissues of 'Luoyang Red' peony.The highest expression was found in flower buds and petals, followed by bracts and leaves, and the least in roots.The expression of Ps FUL1 gene in the petals of different peony cultivars (early flower 'Yingri', midflower 'Luoyang' Hongwei, late flower 'Wang') in 6 different flowering stages was significantly different from that in flowering time, and that in early and late flowering stage, there was a significant difference in the expression of Ps FUL1 gene in the petal of the petal in different flowering stage.The results indicated that Ps FUL1 gene played an important role in regulating the flowering and flowering of peony.
