【摘要】：目的:研究健康促進生活方式和煙堿型乙酰膽堿受體亞單位a5(CHRNA5)-rs 17486278位點基因多態(tài)與肺癌發(fā)病風險的關(guān)系。方法:以2014年12月至2015年12月唐山市工人醫(yī)院確診的216肺癌例患者為病例組,按照年齡、性別、居住地等配對條件選取639例健康體檢人群為對照組。采用聚合酶鏈反應-限制性片段長度多態(tài)性方法(PCR-RFLP)檢測CHRNA5-rs 17486278位點基因多態(tài),結(jié)合兩組人群的健康資料,分析健康促進生活方式和遺傳因素與肺癌發(fā)病風險的關(guān)系。結(jié)果:Logistic回歸顯示健康促進生活方式六個維度中的壓力管理與肺癌發(fā)生有關(guān),OR值為1.770(95%CI:1.262~2.483,P=0.001);病例組和對照組間CHRNA5-rs 17486278等位基因分布頻率之間比較,差異有統(tǒng)計學意義(X~2=10.067,P=0.002);壓力管理與吸煙指數(shù)、吸煙指數(shù)與CHRNA5-rs 17486278位點基因型對肺癌發(fā)生的交互作用,差異有統(tǒng)計學意義(X~2交互作用=4.86,P交互作用=0.034;X~2交互作用=8.62,P交互作用=0.003)。結(jié)論:健康促進生活方式和CHRNA5-rs 17486278位點基因多態(tài)存在協(xié)同作用,增加肺癌發(fā)生的風險。
[Abstract]:Objective: to study the relationship between the health promoting lifestyle and the polymorphism of A5CHRNA5- rs 17486278 gene in the nicotinic acetylcholine receptor subunit. Methods: from December 2014 to December 2015, 216 lungs were diagnosed in Tangshan workers' hospital. Cancer patients as case group, CHRNA5-rs 17486278 locus polymorphism was detected by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method, and the health data of the two groups were analyzed according to the matched conditions of age, sex and residence. The relationship between health promotion lifestyle and genetic factors and lung cancer risk was analyzed. Results: the OR value of stress management in six dimensions of health promotion lifestyle was 1.77095 CI: 1.2622.483P 0.001; case group and control group. The frequency of CHRNA5-rs 17486278 allele distribution was compared. The difference was statistically significant (P < 0.05). The interaction between stress management and smoking index, smoking index and genotype of CHRNA5-rs 17486278 locus on the occurrence of lung cancer. The difference was statistically significant (P < 0.05). Conclusion: there is a synergistic effect between health promoting lifestyle and polymorphism of CHRNA5-rs 17486278 locus gene, which increases the risk of lung cancer.
