水稻HSP90基因家族功能研究

【摘要】:隨著近年來極端天氣的出現(xiàn)愈加頻繁,高溫、低溫、干旱及土壤鹽漬化等逆境脅迫對(duì)作物的生長(zhǎng)、發(fā)育、產(chǎn)量的影響也越發(fā)凸顯。研究作物耐逆機(jī)制,培育耐逆品種成為當(dāng)前亟待解決的問題。HSP90家族蛋白是一類豐富的分子伴侶蛋白,并廣泛存在于真核生物中的保守蛋白。HSP90家族蛋白的主要功能主要是協(xié)助其它蛋白的正確折疊,并在逆境應(yīng)答網(wǎng)絡(luò)中發(fā)揮重要作用。本實(shí)驗(yàn)中利用生物信息學(xué)的工具,發(fā)現(xiàn)并分析了日本晴中HSP90家族的9個(gè)OsHSP90蛋白(OsHSP90-1至OsHSP90-9).通過序列比對(duì),我們?cè)谒綡SP90家族蛋白中確定了三個(gè)基序。通過進(jìn)化樹分析,將水稻HSP90家族蛋白分為三簇(Ⅰ,Ⅱ,Ⅲ)。本實(shí)驗(yàn)在水稻10個(gè)組織中對(duì)這9個(gè)OsHSP90蛋白基因進(jìn)行了表達(dá)量分析。結(jié)果表明在10個(gè)部位的組織中OsHSP90-4、OsHSP90-6和OsHSP90-7有較高的表達(dá)水平,而OsHSP90-5和OsHSP90-8的表達(dá)水平非常低。OsHSP90-1只特異性的在開花14天后的胚中表達(dá)。據(jù)文獻(xiàn)報(bào)道,在熱激和其它逆境脅迫下,熱激蛋白的表達(dá)會(huì)受到正向調(diào)控。但在本研究中,不同逆境脅迫條件下,水稻OsHSP90家族蛋白的表達(dá)模式卻是各式各樣的。在干旱、高鹽、低溫、熱激脅迫下OsHSP90-2和OsHSP90-4的表達(dá)受到了正向調(diào)控,在高鹽、干旱的脅迫下OsHSP90-3和OsHSP90-5的表達(dá)受到了負(fù)向調(diào)控。在干旱脅迫下OsHSP90-7和OsHSP90-9受到負(fù)向調(diào)控。與對(duì)照相比,在多種逆境脅迫下OsHSP90-6的表達(dá)量基本保持不變。我們分別用OsHSP90-2和OsHSP90-4轉(zhuǎn)化大腸桿菌,均能提高轉(zhuǎn)化菌株的細(xì)胞活性并提高對(duì)高溫、高鹽、干旱逆境環(huán)境的耐受力。我們分別用OsHsP90-2和OSHsP90-4轉(zhuǎn)化水稻,但只有OsHSP90-4的水稻轉(zhuǎn)化子對(duì)高溫的耐受力有明顯提高并增強(qiáng)水稻轉(zhuǎn)化子對(duì)ABA的敏感性。這些結(jié)果為進(jìn)一步研究水稻OsHSP90基因家族提供了新的視角。
