水牛IGF1基因多態(tài)性與其生長性狀、血清濃度以及IGF1和IGF1R基因表達的相關性研究(英文)

【摘要】:目的:探討胰島素樣生長因子1(insulin-like growth factor1,IGF1)基因單核苷酸多態(tài)性(SNP)與埃及水牛生長性狀的關聯(lián)性。創(chuàng)新點:發(fā)現(xiàn)IGF1基因多態(tài)性對埃及水牛的生長性狀、血液生化指標和基因表達有顯著影響,并為埃及水牛的選育提供重要的分子遺傳標記。方法:以200頭埃及水牛為試驗對象,對其IGF1基因的多態(tài)性進行檢測,并分析其與生長性狀、血液生理生化指標和基因表達的相關性。結論:在水牛IGF1基因多態(tài)性分析中,發(fā)現(xiàn)兩個新的SNP位點(G64A和G280A)分別分布在外顯子1和外顯子4的非編碼區(qū)。不同基因型的統(tǒng)計分析表明,G64A和G280A位點的GG基因型水牛個體分別在3~6月齡和6~9月齡的體重(BW)和平均日增重(ADG)均顯著高于其他基因型(P0.05)。這兩個SNPs位點的組合基因型產(chǎn)生了三種單倍體GG/GG、AG/AG和AA/AA。在3~12月齡的水牛個體中,單倍體基因型與BW和ADG存在顯著關聯(lián)(P0.0001)。純合的GG/GG單倍體基因型水牛生長性能優(yōu)于其他水牛。兩個SNP位點與半腱肌中IGF1和IGF1R的mRNA水平以及IGF1血清濃度水平相關。此外,GG/GG單倍體水牛表現(xiàn)出較高的mRNA和血清濃度水平。綜上所述,這兩種SNP位點G64A和G280A可作為埃及水牛生長性狀選育的重要分子遺傳標記。
[Abstract]:Objective: To investigate the effects of insulin-like growth factor 1 (insulin-like growth, factor1, IGF1) gene single nucleotide polymorphisms (SNP) associated with growth traits. The Egyptian buffalo Innovation: IGF1 gene polymorphisms on growth traits of Egypt Buffalo, blood biochemical index and gene expression have a significant effect, provide an important molecular genetic markers and breeding of Egyptian buffalo. Methods: 200 head of buffalo Egypt as test object, detect the polymorphism of the IGF1 gene, and to analyze its correlation with growth traits, physiological and biochemical indexes of blood and gene expression. Conclusion: in the analysis of buffalo IGF1 gene polymorphism in the discovery of two new SNP loci (G64A and G280A) respectively in non encoding region of exon 1 and exon 4. Statistical analysis showed that different genotypes, G64A and G280A loci GG genotype individual buffalo respectively in 3~6 months and 6~9 months weight (BW) And the average daily gain (ADG) were significantly higher than those of other genotypes (P0.05). The combined genotypes of two SNPs loci produced three haploid GG/GG, individual buffalo at the age of 3~12 months, AG/AG and AA/AA., BW and ADG haploid genotype and significant association (P0.0001). The GG/GG genotype of haploid buffalo pure growth performance is better than other buffalo. IGF1 and IGF1R two SNP loci and semitendinosus muscle in mRNA level and IGF1 level of serum. In addition, the buffalo GG/GG haplotype exhibited higher mRNA and serum level. To sum up, the two SNP loci G64A and G280A can be used as an important molecular growth traits of Egypt buffalo the selection of genetic markers.
