【摘要】:膨脹素是植物細胞特有的一種細胞壁松弛蛋白,在很大程度上決定著植物細胞的生長速率。α-膨脹素(α-expansin,EXPA)和β-膨脹素(β-expansin,EXPB)是植物膨脹素家族中兩個較大的亞家族。水稻(Oryza sativa)中至少有34個α-膨脹素基因。本研究通過轉(zhuǎn)基因技術(shù),研究了一個水稻根特異性表達的α-膨脹素基因Os EXPA8對水稻懸浮細胞的細胞周期和細胞大小的影響。利用流式細胞儀,分析不同培養(yǎng)時期野生型和轉(zhuǎn)基因型細胞周期的差異,結(jié)果發(fā)現(xiàn),35S::Os EXPA8(過表達)轉(zhuǎn)基因懸浮細胞大部分處于G2/M期,而Os EXPA8-RNAi(knock-down)轉(zhuǎn)基因懸浮細胞多處在G1期;利用血球計數(shù)板測定細胞數(shù)量,結(jié)果表明,35S::Os EXPA8轉(zhuǎn)基因細胞數(shù)量野生型細胞數(shù)量Os EXPA8-RNAi轉(zhuǎn)基因細胞數(shù)量;利用顯微鏡測定不同培養(yǎng)時期的細胞大小,發(fā)現(xiàn)35S::Os EXPA8轉(zhuǎn)基因細胞大小野生型細胞大小Os EXPA8-RNAi轉(zhuǎn)基因細胞大小;測定愈傷組織塊大小,發(fā)現(xiàn)35S::Os EXPA8轉(zhuǎn)基因愈傷組織塊大小野生型愈傷組織塊大小Os EXPA8-RNAi轉(zhuǎn)基因愈傷組織塊大小。本研究結(jié)果表明,Os EXPA8能夠促進水稻懸浮細胞的分裂和生長,從而增加細胞數(shù)量,增大細胞體積,對水稻懸浮細胞的生長具有調(diào)控作用。該研究將有助于更好地了解植物細胞壁的松弛過程,從而深入地認識植物的生長發(fā)育進程。同時,膨脹素作為一種細胞壁松弛因子在農(nóng)業(yè)上具有潛在的生物工程應(yīng)用價值。
[Abstract]:Expansin is a cell wall loosening proteins in plant cell specific, largely determines the growth rate of plant cells. Alpha expansin (alpha -expansin, EXPA) and beta expansin (beta -expansin, EXPB) is a plant expansion in two large family of rice (Oryza sativa subfamily.) at least 34 alpha expansin genes. This study by transgenic technology, studies on an expression of rice root specific alpha expansion effect of Os gene on cell cycle of EXPA8 in rice suspension cell and cell size. Using flow cytometry, analysis of differences in different culture period of wild type and transgenic type the cell cycle was found: 35S: Os EXPA8 (expression) of most transgenic suspension cells in G2/M phase, and Os EXPA8-RNAi (knock-down) transgenic suspension cells in G1 phase; Determination of cell number using hemocytometer.
