柱前衍生高效液相色譜法測定二乙醇胺脫氫產(chǎn)物亞氨基二乙酸和甘氨酸

【摘要】:以對甲苯磺酰氯(PTSC)為衍生劑,建立了柱前衍生高效液相色譜(HPLC)測定二乙醇胺脫氫產(chǎn)物中亞氨基二乙酸(IDA)和甘氨酸(Gly)含量的分析方法。IDA和Gly與衍生劑在堿性(pH 11)條件下于45℃反應(yīng)15 min,進(jìn)行柱前衍生,并利用高效液相色譜-質(zhì)譜對衍生產(chǎn)物進(jìn)行定性分析。衍生化產(chǎn)物采用VP-ODS色譜柱(200 mm×4.6 mm,5μm)分離,以0.03 mol/L醋酸銨溶液(pH 5.5)為流動(dòng)相A、乙腈為流動(dòng)相B(體積比為87∶13),進(jìn)行等度洗脫,流速為1 mL/min,并采用配有紫外檢測器的高效液相色譜儀測定,檢測波長為235 nm。該法在IDA質(zhì)量濃度為900~2 100 mg/L、Gly質(zhì)量濃度為20~100 mg/L的范圍內(nèi)線性關(guān)系良好,相關(guān)系數(shù)(R2)均大于0.999。IDA和Gly的檢出限(LOD)分別為0.089 7 mg/L和0.026 2 mg/L,加標(biāo)回收率分別為98.7%~99.3%和98.0%~99.5%,相對標(biāo)準(zhǔn)偏差(RSD)分別為0.89%~1.23%和0.95%~1.11%(n=3)。該法具有反應(yīng)條件溫和、準(zhǔn)確性高的特點(diǎn),可用于工藝生產(chǎn)中IDA和Gly含量的測定。
