基于RRLC-DAD-ESI-Q-TOF-MS技術(shù)的山東金銀花多指標(biāo)定量指紋圖譜分析

【摘要】:目的:運(yùn)用RRLC-DAD-ESI-Q-TOF-MS技術(shù)分析和測(cè)定金銀花中主要化學(xué)成分,建立山東金銀花多指標(biāo)定量指紋圖譜。方法:采用RRLC-DAD-ESI-Q-TOF-MS技術(shù)對(duì)金銀花提取物進(jìn)行快速分析,根據(jù)獲得的精確相對(duì)分子質(zhì)量和二級(jí)質(zhì)譜碎片信息推斷化合物分子式,結(jié)合數(shù)據(jù)庫(kù)和文獻(xiàn)對(duì)金銀花中化學(xué)成分進(jìn)行鑒別,采用RRLC-DAD法分析測(cè)定山東金銀花、河南金銀花和湖南山銀花中主要化學(xué)成分含量,在此基礎(chǔ)上建立了山東金銀花多指標(biāo)定量指紋圖譜。結(jié)果:從山東金銀花中檢測(cè)到51種化學(xué)成分,鑒定了其中32種化學(xué)成分;對(duì)比測(cè)定了山東金銀花、河南金銀花和湖南山銀花中10種主要化學(xué)成分(馬錢酸、綠原酸、咖啡酸、馬錢苷、斷氧化馬錢苷、蘆丁、金桃苷、木犀草苷、異綠原酸A和異綠原酸C)的含量;采用指紋圖譜結(jié)合相似度分析可以實(shí)現(xiàn)不同品種金銀花和湖南山銀花的正確區(qū)分,所得結(jié)果與含量測(cè)定結(jié)果一致。結(jié)論:RRLC-DAD-ESIQ-TOF-MS技術(shù)分析速度快、靈敏度高、能夠提供化合物及其碎片離子的精確質(zhì)量信息,可以快速準(zhǔn)確地鑒別和測(cè)定金銀花中的化學(xué)成分。在此基礎(chǔ)上建立的山東道地金銀花的多指標(biāo)定量指紋圖譜,為山東金銀花質(zhì)量評(píng)價(jià)研究提供了技術(shù)支持。
[Abstract]:Objective: to analyze and determine the main chemical constituents in Lonicera japonica by RRLC-DAD-ESI-Q-TOF-MS, and to establish quantitative fingerprint of Flos Lonicerae in Shandong Province. Methods: the extract of Flos Lonicerae was analyzed quickly by RRLC-DAD-ESI-Q-TOF-MS, and the molecular formula of the compound was deduced according to the accurate molecular weight and the information of second order mass spectrometry. The chemical constituents of Flos Lonicerae in Shandong Province, Henan Province and Hunan Province were determined by RRLC-DAD method. On this basis, the quantitative fingerprint of honeysuckle in Shandong was established. Results: 51 chemical constituents were detected from Flos Lonicerae in Shandong Province and 32 of them were identified. Ten main chemical constituents (equinic acid, Lv Yuan acid, caffeic acid, riganoside, Rutin, hyperin, luteolin) in Honeysuckle, Henan Flos Lonicerae and Honeysuckle in Hunan Province were determined, and the chemical constituents of Flos Lonicerae, Honeysuckle, Honeysuckle and Honeysuckle in Hunan were determined. Contents of isoLv Yuan acid A and isob_person1# acid C; Fingerprint and similarity analysis can be used to distinguish different varieties of Flos Lonicerae from Honeysuckle in Hunan Province. The results are consistent with the results of content determination. Conclusion: RRLC-DAD-ESIQ-TOF-MS can provide accurate mass information of compounds and their fragments, and can be used to identify and determine the chemical constituents of Flos Lonicerae. On this basis, the quantitative fingerprint of Lonicera japonica in Shandong Province was established, which provided technical support for the quality evaluation of Lonicera japonica in Shandong.
