UPLC法測定香蕉、皇帝蕉皮及其果肉中羽扇豆酮含量

【摘要】:目的:建立超高效液相色譜法分離和測定不同收集期的香蕉和皇帝蕉皮及其果肉中羽扇豆酮含量的方法,并優(yōu)化提取條件。方法:采用甲醇回流提取法對2種蕉皮及其果肉的羽扇豆酮進行提取分離,減壓濃縮,選用Waters ACQUITY UPLC BEH C18色譜柱(2.1 mm×50 mm,1.7μm),流動相為乙腈-水(85∶15,V/V)溶液,流速0.3 mL/min,柱溫30℃,檢測波長206 nm。結(jié)果:香蕉和皇帝蕉果肉中均未檢測出羽扇豆酮,但2種蕉皮中有較高含量的羽扇豆酮,并且在16.70~59.96 mg/100 mL質(zhì)量濃度范圍內(nèi)有良好線性關(guān)系(r=0.999 6),羽扇豆酮平均回收率為99.00%,相對標(biāo)準(zhǔn)偏差為3.0%。結(jié)論:該法回收率高、重復(fù)性好,且具有專屬性強、準(zhǔn)確性高和簡便快捷等特點,適用于香蕉、皇帝蕉皮中羽扇豆酮的含量測定。
