【摘要】：Daphlongamine E是郝小江小組于2009年從長序虎皮楠葉子中分離得到的一種Calyciphylline A類虎皮楠生物堿。研究表明,Daphlongamine E中含特殊的氮雜[3.3.1]橋環(huán)體系和[6/6/5/7/5]五環(huán)剛性骨架以及六個(gè)手性中心,其復(fù)雜的剛性結(jié)構(gòu)使得對它的全合成研究成為一項(xiàng)極具挑戰(zhàn)性的工作。在本文中,我們從手性化合物(-)-2-1出發(fā),以金催化烯醇硅醚的環(huán)化反應(yīng),有機(jī)膦催化的[3+2]環(huán)加成反應(yīng),銥(I)催化的不對稱氫化反應(yīng),酰基芳�；^氧化物的重排反應(yīng)以及分子內(nèi)的aldol反應(yīng)為關(guān)鍵步,總共17步完成了Daphlongamine E的不對稱五環(huán)骨架的構(gòu)建,為Daphlongamine E的不對稱全合成奠定了基礎(chǔ)。
[Abstract]:Daphlongamine E is a Calyciphylline A alkaloid isolated from the leaves of Cymbidium longensis by Hao Xiaojiang team in 2009. It is shown that Daphlongamine E contains a special [3.3.1] bridged ring system and [6 / 6 / 5 / 7 / 5] pentacyclic rigid skeleton and six chiral centers. Its complex rigid structure makes it a challenging task to study its full synthesis. In this paper, starting from chiral compound (-) -2-1, we use gold to catalyze the cyclization of enol silyl ether, [32] cycloaddition reaction catalyzed by organic phosphine, and asymmetric hydrogenation reaction catalyzed by iridium (I). The rearrangement of acyl aryl peroxide and the intramolecular aldol reaction were the key steps. The asymmetric pentacyclic skeleton of Daphlongamine E was constructed in 17 steps, which laid the foundation for asymmetric synthesis of Daphlongamine E.
