【摘要】：在利用IR光譜原位監(jiān)測酶促反應(yīng)的過程中,反應(yīng)物與底物紅外特征吸收峰會隨反應(yīng)進(jìn)程發(fā)生變化,因此可根據(jù)單位時間內(nèi)特征峰吸收值的變化量來反映酶的催化效率。然而,該方法在實(shí)際應(yīng)用過程中還存在諸多的局限性有待改進(jìn)。例如,峰位選取不準(zhǔn)確和譜帶嚴(yán)重重疊無法分辨/分離等都將給酶活力的測試結(jié)果帶來偏差。二維相關(guān)分析與IR光譜的結(jié)合使用將在一定程度上改進(jìn)上述問題,我們將在本文中以具體事例分別闡述。
[Abstract]:In the process of in situ monitoring the enzymatic reaction by IR spectroscopy, the infrared characteristic absorption summit of reactants and substrates changes with the reaction process, so the catalytic efficiency of the enzyme can be reflected according to the change of the absorption value of the characteristic peak in unit time. However, there are still many limitations in the practical application of this method. For example, the inaccuracy of peak selection and the serious overlap of spectrum bands can not distinguish / separate the enzyme activity, which will bring deviation to the test results of enzyme activity. The combination of two dimensional correlation analysis and IR spectra will improve the above problems to some extent. We will give some examples in this paper.
