離子色譜法測定不同產(chǎn)地葛根多糖中的單糖組成

【摘要】:建立了葛根中巖藻糖、鼠李糖、阿拉伯糖、半乳糖、葡萄糖、甘露糖、木糖、核糖、甘露醇、木糖醇、果糖等11種單糖的離子色譜方法,并應(yīng)用此方法分析了不同產(chǎn)地葛根多糖中單糖組成及含量。葛根多糖經(jīng)超聲提取,Box-Behnken響應(yīng)面優(yōu)化超聲提取條件,三氟乙酸水解,以Carbo Pac PA20(150 mm×3 mm)色譜柱分離,20 mmol/L NaOH溶液等度洗脫。結(jié)果表明,11種單糖的定量限為0.03~0.07 mg/kg,在0.2~5 mg/L線性范圍內(nèi)線性關(guān)系良好(r~20.9990),加標(biāo)回收率為80.3%~112.4%。經(jīng)Box-Behnken響應(yīng)面優(yōu)化后的最佳提取工藝為液料比28:1(mL/g),超聲時(shí)間44 min,超聲溫度65℃。經(jīng)離子色譜方法測定不同產(chǎn)地的葛根樣品中均含有巖藻糖、鼠李糖、葡萄糖,其中葡萄糖的含量最高含量在114.3~622.2 mg/g之間。方法可為葛根質(zhì)量評價(jià)提供技術(shù)支撐。
[Abstract]:A method for ion chromatography of 11 monosaccharides, such as fucose, rhamnose, arabinose, galactose, glucose, mannose, xylose, ribose, mannitol, xylitol and fructose, was established. The composition and content of monosaccharide in Pueraria lobata polysaccharides from different areas were analyzed by this method. The ultrasonic extraction conditions of Pueraria polysaccharide were optimized by ultrasonic extraction Box-Behnken response surface, trifluoroacetic acid was hydrolyzed, and iso-elution of 20 mmol/L NaOH solution was separated by Carbo Pac PA20(150 mm 脳 3 mm chromatographic column. The results showed that the quantitative limit of 11 monosaccharides was 0.03 ~ 0.07 mg / kg, the linear range was 0.2 ~ 5 mg/L and the recovery rate was 80.3% ~ 112.4%.
