嗜鹽桿菌HSQAY1對中肋骨條藻的溶藻物質特性

【摘要】:研究嗜鹽桿菌菌株(Halobacillus sp.HSQAY1)發(fā)酵液在不同p H、溫度和反復凍融條件下對中肋骨條藻(Skeletonema costatum)溶藻活性的環(huán)境穩(wěn)定性;并采用乙醇沉淀、硫酸銨沉淀、超濾和聚丙烯酰胺凝膠電泳(SDS-PAGE)等方法對溶藻物質的理化特性進行了初步探討和分析。結果表明,該菌株產生的溶藻物質在強酸性條件下(p H=3)喪失活性,p H5~11時的溶藻率在83.4%~98.4%之間;30~110℃時溶藻率在71.7%~94.5%之間,表現出一定的酸堿穩(wěn)定性和熱穩(wěn)定性,且不受反復凍融(-20℃)影響。溶藻活性物質具有被乙醇和硫酸銨沉淀的特性,其活性組分是多種分子質量10k Da的蛋白類物質,溶藻活性蛋白粗提物的SDS-PAGE電泳顯示,在15~50 k Da間有3條明顯的特異蛋白條帶。
[Abstract]:The environmental stability of algae-lytic activity of Halobacillus sp. HSQAY1 fermentation broth against Skeletonema costatum under different pH, temperature and repeated freeze-thaw conditions was studied, and ethanol precipitation and ammonium sulfate precipitation were used to study the environmental stability of the lysogenic activity of Halobacillus sp. HSQAY1 fermentation broth against Skeletonema costatum under different pH, temperature and repeated freeze-thaw conditions. The physicochemical properties of algae-lytic substances were preliminarily discussed and analyzed by ultrafiltration and SDS-PAGE-based gel electrophoresis. The algae-soluble substance produced by the strain was inactivated under strong acidic conditions. The algae-lysing rate of the strain was between 83.4- 98.4% and 30.110 鈩,
