油菜籽蛋白質(zhì)膜的制備與性能研究

【摘要】:菜籽蛋白是一類優(yōu)質(zhì)的植物蛋白資源,含有大量的分子間作用力,范德華力,二硫鍵以及疏水相互作用使得菜籽蛋白具有成膜的潛質(zhì)。本文通過使用瓊脂復(fù)配制備了性能優(yōu)良的菜籽蛋白質(zhì)薄膜,并對菜籽蛋白的提取過程以及最佳的成膜條件進行了探究,并對其在包裝領(lǐng)域的應(yīng)用上進行了初步的探究。以菜籽粕為原料,采用酸提堿沉淀的方法提取菜籽蛋白,并建立了利用菜籽蛋白結(jié)合色素自身顯色的方法來測定菜籽蛋白的含量。以不同濃度的菜籽蛋白溶液繪制標(biāo)準(zhǔn)曲線,并用凱氏定氮法標(biāo)定曲線,其1.0-3.0mg/m L范圍內(nèi)線性關(guān)系良好,線性相關(guān)系數(shù)R2大于0.999,平均回收率為99.1%,精密度,重現(xiàn)性良好,適用于大批量同品種菜籽蛋白的快速檢測。以菜籽提取蛋白制備菜籽蛋白質(zhì)薄膜,并對薄膜的性質(zhì)進行了研究,結(jié)果顯示,菜籽蛋白成膜溶液最佳的處理條件為甘油含量20%,菜籽蛋白含量為30%,處理溫度50℃,p H10.0,紫外輻射處理24h。在此條件下,薄膜的抗拉強度為3.5MPa,斷裂伸長率為20%。采用十二烷基硫酸鈉改性的埃洛石納米管對薄膜進行改性研究,結(jié)果發(fā)現(xiàn),最佳的改性劑用量為3%,在此條件下薄膜的抗拉強度提高約20%,但是其斷裂伸長率則降低約10%。微觀結(jié)果測試顯示,薄膜的層狀結(jié)構(gòu)已經(jīng)改變?yōu)閱螌拥谋∧?并且改性埃洛石納米管通過物理吸附與化學(xué)交聯(lián)的方式共同改善薄膜的性質(zhì)。菜籽蛋白質(zhì)與瓊脂共混膜的最佳制備條件為菜籽蛋白與瓊脂比為3:7,其他條件與制備菜籽蛋白膜相同,此時薄膜的抗拉強度和斷裂伸長率分別為35MPa和10%,較單一的菜籽蛋白膜有大幅的提升;同時共混膜的透明度以及阻隔性能有了大幅的改善。結(jié)果說明,共混是改善蛋白質(zhì)薄膜的性能的有效手段。薄膜的微觀結(jié)構(gòu)顯示,共混后的菜籽蛋白膜具有更加致密的結(jié)構(gòu),這也說明了共混是提高蛋白質(zhì)薄膜性能的有效手段。在對菜籽蛋白共混膜的改性研究中發(fā)現(xiàn),使用對菜籽蛋白膜的安全性做了初步的探究,結(jié)果說明,菜籽蛋白有毒物質(zhì)含量低于相關(guān)標(biāo)準(zhǔn),薄膜在高溫下不會揮發(fā)出有毒有害物質(zhì)。
[Abstract]:Rapeseed protein (rapeseed protein) is a kind of high quality plant protein resource, which contains a lot of intermolecular force, van der Waals force, disulfide bond and hydrophobic interaction, which makes rapeseed protein have the potential of film formation. In this paper, rapeseed protein thin films with excellent properties were prepared by using Agar, the extraction process and the optimum conditions of film formation were studied, and the application of rapeseed protein in packaging field was preliminarily explored. Rapeseed protein was extracted from rapeseed meal by acid extraction and alkali precipitation. The method of coloring rapeseed protein binding pigment was established to determine the content of rapeseed protein. The standard curve was drawn with different concentrations of rapeseed protein solution and calibrated by Kjeldahl method. The linear correlation coefficient R 2 was greater than 0.999 9, the average recovery was 99.1%, the precision and reproducibility were good, the linear correlation coefficient R 2 was greater than 0.999 9, and the calibration curve was calibrated by Kjeldahl method. It is suitable for rapid detection of rapeseed protein of the same variety in large quantities. Rapeseed protein films were prepared by extracting protein from rapeseed, and the properties of the films were studied.
