亞麻刺盤孢表面磁納米修飾及其羥化去氫表雄酮

【摘要】:以陽離子聚電解質(zhì)聚烯丙基胺鹽酸鹽(PAAH)對共沉淀法制備的Fe_3O_4磁性納米顆粒進(jìn)行包裹,再以此對亞麻刺盤孢Colletotrichum lini ST-1進(jìn)行表面修飾,制備具有磁性的菌體復(fù)合物。通過SEM、XRD、VSM和zeta電位分析表明,PAAH修飾的Fe_3O_4能夠穩(wěn)定均一地結(jié)合在C.lini ST-1菌體表面,形成的磁性復(fù)合物具有超順磁性,可利用外加磁場對菌體進(jìn)行回收。本研究利用該磁性復(fù)合物對去氫表雄酮(DHEA)進(jìn)行生物羥化,制備三羥基雄甾烯酮(7α,15α-di OH-DHEA),通過回收菌體完成了4個(gè)批次的轉(zhuǎn)化,產(chǎn)物7α,15α-di OH-DHEA的濃度為10.83g/L,約為游離菌體轉(zhuǎn)化效果的3倍。
[Abstract]:The Fe_3O_4 magnetic nanoparticles prepared by coprecipitation method were encapsulated with polyallylamine hydrochloride (PAAH), a cationic polyelectrolyte, and the surface of Colletotrichum lini ST-1 was modified by this method. The magnetic complex was prepared. The results of SEMS-XRDS-VSM and zeta potential analysis showed that the modified Fe_3O_4 could bind stably and homogeneously to the surface of C.lini ST-1, and the resulting complex had superparamagnetic properties. In this study, the magnetic complex was used for biohydroxylation of dehydroepiandrosterone (DHEAA) to prepare trihydroxyandrostenone 7 偽 -di OH-DHEAA. The concentration of the product was 10.83 g / L, which was about 3 times of that of free cell transformation.
