長期遞增負荷運動對大鼠股骨OPG和RANKL蛋白表達的影響

【摘要】:目的:觀察長期遞增負荷運動對大鼠股骨OPG、RANKL蛋白表達以及BMD的影響效應。方法:130只3月齡SD雌性大鼠,適應性訓練后隨機分為運動組和對照組。運動組分別進行為期4、9、11、13、15、17 W的遞增負荷跑臺運動(E_4、E_9、E_(11)、E_(13)、E_(15)、E_(17)),并設立同期對照組(C_4、C_9、C_(11)、C_(13)、C_(15)、C_(17))和基礎對照組(BC),每組10只。于末次運動后12~24 h,以DXA測定大鼠全身BMD,用Western Blot方法測定股骨OPG和RANKL的蛋白表達。結(jié)果:在17 W遞增負荷跑臺運動過程中,運動組大鼠OPG表達及OPG/RANKL比值呈現(xiàn)先升高趨勢,于第9 W達到峰值,隨后呈下降的趨勢,于17 W降至最低(E_(17)vs.C17,P0.05,ES=2.66;P0.05,ES=2.65)。運動組可溶性RANKL(sRANKL)、膜結(jié)合型RANKL(mRANKL)及總RANKL表達呈現(xiàn)先下降趨勢,于第9 W或11 W時降至最低,隨后呈逐漸升高的趨勢。在17 W運動過程中,運動組大鼠全身BMD先表現(xiàn)為增加趨勢,于第9W達到峰值(E_9vs.C_9,P0.05,ES=0.85),隨后呈下降趨勢,于17 W降至最低(E_(17)vs.C_(17),P0.01,效果量=1.49)。結(jié)論:長期遞增負荷運動過程中,前9 W適量運動可上調(diào)OPG表達、下調(diào)RANKL表達,抑制骨吸收,促進BMD增加;后期過量運動則抑制OPG表達、促進RANKL表達,骨吸收作用增強,導致BMD下降。
[Abstract]:Aim: to observe the effect of long-term incremental exercise on the expression of OPG,RANKL protein and BMD in rat femur. Methods: 130 3-month-old female SD rats were randomly divided into exercise group and control group after adaptive training. The exercise group was divided into 4, 9, 11, 13, 15, 17 W incremental load races (E / 4, E _ (9), E _ (11), E _ (13), E _ (15), E _ (17),) and the control group (C ~ (4), C ~ (9), C _ (17), C _ (11), E _ (13), E _ (15), E _ (17),). C _ (11), C _ (13), C _ (15), C _ (17) and basal control group (BC),) were 10 rats in each group. At 12 h after the last exercise, the protein expression of OPG and RANKL in femur was measured by Western Blot method in the whole body of BMD, of rats. DXA was used to measure the expression of OPG and RANKL in the whole body of rats. Results: the expression of OPG and the ratio of OPG/RANKL in the exercise group increased at first, reached the peak at the 9th week, then decreased to the lowest level at 17 W (E17 vs.C17,). P0.05, ESL 2.66; (P 0.05, ESL 2.65). In the exercise group, the expression of soluble RANKL (sRANKL), membrane-bound RANKL (mRANKL) and total RANKL decreased first, decreased to the lowest at 9 W or 11 W, and then increased gradually. During 17 W exercise, the whole body BMD of rats in the exercise group showed an increasing trend at first and reached a peak at the 9th week (E = 9 vs. C = 9, P 0.05, es = 0.85), and then showed a decreasing trend, while the whole body BMD of the exercise group showed an increasing trend at first and reached a peak at the 9th week. It decreased to the lowest level at 17 W (E17 vs.C_ (17), P0.01, effect = 1.49). Conclusion: during long-term incremental exercise, moderate exercise in the first 9 W can up-regulate the expression of OPG, down-regulate the expression of RANKL, inhibit bone resorption and promote the increase of BMD. Excessive exercise inhibited the expression of OPG, promoted the expression of RANKL and increased the bone resorption, which resulted in the decrease of BMD.
