過(guò)氧化氫酶基因多態(tài)性在運(yùn)動(dòng)員選材中應(yīng)用的可行性研究

【摘要】:研究目的:通過(guò)分析過(guò)氧化氫基因(CAT)多態(tài)性在中國(guó)漢族運(yùn)動(dòng)員和對(duì)照人群中的分布特征及該位點(diǎn)多態(tài)性與運(yùn)動(dòng)員選材用生理生化指標(biāo)的關(guān)聯(lián),以期從遺傳學(xué)角度探尋與運(yùn)動(dòng)能力相關(guān)的基因標(biāo)記。研究對(duì)象與方法:本研究選取了CAT基因SNP/A-89T位點(diǎn)。采用酶切和測(cè)序的方法分析CAT基因SNP/A-89T多態(tài)性在92名中國(guó)漢族運(yùn)動(dòng)員和110名漢族普通對(duì)照組中的分布特征。同時(shí)測(cè)試運(yùn)動(dòng)員的體成分最大攝氧量、血尿素、肌酸激酶及血象指標(biāo),試圖探尋CAT基因SNP/A-89T作為運(yùn)動(dòng)員選材用基因指標(biāo)的可行性。研究結(jié)果:(1)CAT基因SNP/A-89T在漢族普通人群中基因型分布為T(mén)T型最少,AT型次之,AA型最多。這與國(guó)內(nèi)其它地區(qū)漢族人群間分布頻率未見(jiàn)顯著性差異,但與日本人群以及南美人群的基因型分布頻率存在顯著性差異。提示CAT基因SNP/A-89T位點(diǎn)在國(guó)內(nèi)各地域間分布特征不存在差異,但可作為種族差異的基因標(biāo)記。(2)CAT基因SNP/A-89T位點(diǎn)在漢族運(yùn)動(dòng)員和對(duì)照組中基因型頻率和等位基因頻率均無(wú)性別地域差異。將運(yùn)動(dòng)員按照級(jí)別分組后發(fā)現(xiàn),女子一級(jí)運(yùn)動(dòng)員攜A等位基因的分布頻率明顯高于攜T等位基因的頻率。按照供能系統(tǒng)分組,發(fā)現(xiàn)有氧項(xiàng)目女子運(yùn)動(dòng)員(AT+TT)型的分布頻率顯著高于AA型,攜T等位基因的頻率顯著高于A等位基因。(3)CAT基因SNP/A-89T與選材用部分生理生化指標(biāo)有關(guān)聯(lián)。在有氧項(xiàng)目中,發(fā)現(xiàn)男子運(yùn)動(dòng)員AA型去脂體重顯著高于(AT+TT)型運(yùn)動(dòng)員;男女運(yùn)動(dòng)員攜(AT+TT)型者的血紅蛋白均顯著高于AA型;男子運(yùn)動(dòng)員中攜(AT+TT)型者在VO2max(ml/min/Kg)指標(biāo)顯著高于AA型;女子運(yùn)動(dòng)員攜(AT+TT)型在血尿素指標(biāo)中顯著低于AA型。研究結(jié)論:CAT基因SNP/A-89T攜(AT+TT)型可作為有氧運(yùn)動(dòng)員選材用基因指標(biāo),但還需要加大樣本量進(jìn)一步研究。
[Abstract]:Objective: to analyze the distribution of hydrogen peroxide gene CAT polymorphism in Chinese Han athletes and the association between the polymorphism and physiological and biochemical indexes.The aim is to explore the genetic markers related to motor ability from the perspective of genetics.Methods: the SNP/A-89T locus of CAT gene was selected in this study.The distribution of CAT gene SNP/A-89T polymorphism in 92 Chinese Han athletes and 110 Han normal controls was analyzed by enzyme digestion and sequencing.At the same time, the maximal oxygen uptake, blood urea, creatine kinase and blood image index of athletes were tested to explore the feasibility of using CAT gene SNP/A-89T as a genetic index for selecting athletes.The results showed that the genotype distribution of cat gene SNP/A-89T was the lowest in TT type and second in AT type in Han population.There was no significant difference in distribution frequency between Han population in other regions of China, but there was significant difference in genotype distribution with Japanese population and South American population.It was suggested that there was no difference in the distribution of SNP/A-89T loci of CAT gene among different regions in China, but there was no gender difference in genotype frequency and allele frequency of SNP/A-89T locus of cat gene in Han athletes and controls.The distribution frequency of A allele was obviously higher than that of T allele.According to the energy supply system, it was found that the distribution frequency of AT TTT type in aerobic women athletes was significantly higher than that of AA type, and the frequency of carrying T allele was significantly higher than that of A allele. The SNP/A-89T of catalase gene was correlated with some physiological and biochemical indexes of material selection.In aerobic events, it was found that the body weight of male athletes with AA type fat removal was significantly higher than that of AT type athletes, that of male and female athletes with AT TTT type was significantly higher than that of AA type, that of male athletes with AT TTT type was significantly higher than that of AA type, that of male athletes with AT TTT type was significantly higher than that of AA type.The blood urea index of female athletes was significantly lower than that of AA type.Conclusion the SNP/A-89T / AT TTT type of the 1: cat gene can be used as a genetic index for the selection of aerobic athletes, but there is still a need for further research on the sample size.
