補(bǔ)充β-丙氨酸對(duì)間歇訓(xùn)練大鼠糖代謝指標(biāo)的影響

【摘要】:目的:通過(guò)給間歇運(yùn)動(dòng)訓(xùn)練的大鼠補(bǔ)充p-丙氨酸,觀察大鼠骨骼肌中肌肽含量的變化,探索補(bǔ)充p-丙氨酸對(duì)間歇訓(xùn)練大鼠骨骼肌以及肝臟糖代謝的影響。方法:將40只八周齡的健康雄性SD大鼠,隨機(jī)分成4組(N=10):間歇運(yùn)動(dòng)中等劑量補(bǔ)劑組(A組)、間歇運(yùn)動(dòng)高劑量補(bǔ)劑組(B組)、間歇運(yùn)動(dòng)補(bǔ)水組(C組)、空白對(duì)照組(D組)。運(yùn)動(dòng)方式采用高速間歇訓(xùn)練,跑臺(tái)速度為50 m/min,時(shí)間6 min,休息5 min后,繼續(xù)依此方式共訓(xùn)練3次,每周6天,共4周。補(bǔ)劑采用灌胃方式,中等劑量組補(bǔ)充劑量為1g/kg體重,高劑量組補(bǔ)充劑量為1.5g/kg體重,補(bǔ)水組和空白對(duì)照組則為相同體積的蒸餾水。然后采用鄰苯二甲醛柱前衍生化法測(cè)試腓腸肌中肌肽含量、比色法測(cè)腓腸肌肌糖原含量、比色法測(cè)腓腸肌己糖激酶活性;葡萄糖氧化酶法測(cè)血清中葡萄糖含量;比色法測(cè)肝臟中肝糖原含量以及實(shí)時(shí)熒光定量PCR測(cè)肝臟葡萄糖-6-磷酸酶、糖皮質(zhì)激素受體、糖原合成酶、葡萄糖激酶和胰島素受體的mRNA相對(duì)表達(dá)。結(jié)果:(1)A組大鼠腓腸肌中肌肽含量顯著高于C組(p0.05),B組也顯著高于C組(p0.01);C組大鼠腓腸肌己糖激酶活性顯著低于D組,A組顯著高于B、C兩組(p0.01)。(2)C組的大鼠肝糖原含量顯著高于D組(p0.01),A組顯著低于B、C兩組(p0.05);C組大鼠肝臟中葡糖糖-6-磷酸酶的mRNA相對(duì)表達(dá)顯著高于D組(p0.01);A組大鼠肝臟中糖皮質(zhì)激素受體的mRNA相對(duì)表達(dá)顯著高于B、C兩組(p0.01);C組大鼠肝臟中糖原合成酶的mRNA相對(duì)表達(dá)顯著低于D組(p0.01);A組大鼠胰島素受體的mRNA相對(duì)表達(dá)顯著高于B組(p0.01)。結(jié)論:補(bǔ)充p-丙氨酸可以促進(jìn)間歇訓(xùn)練大鼠肌肉對(duì)血糖的利用,增加肝臟糖原的分解,中等劑量效果更好。
[Abstract]:Objective: to observe the change of carnosine content in skeletal muscle of rats by adding palanine to intermittent exercise training rats. To explore the effect of supplementation of p-alanine on glucose metabolism in skeletal muscle and liver of rats after intermittent training. The rats were randomly divided into 4 groups: group A with moderate dose of intermittent exercise, group B with high dose of intermittent exercise, group C with intermittent exercise, group D with blank control group. The treadmill speed was 50 m / min, time 6 minutes, rest for 5 min, then continued to train 3 times a week, 6 days a week, for 4 weeks. The supplementation was done by gavage, the medium dose group was 1g/kg body weight, and the high dose group was 1.5g/kg body weight. The contents of carnosine in gastrocnemius muscle, glycogen content in gastrocnemius muscle and hexokinase activity in gastrocnemius muscle were measured by pre-column derivatization method, colorimetric method and colorimetric method respectively. Glucose content in serum was measured by glucose oxidase method, liver glycogen content was measured by colorimetric method, glucose-6-phosphatase, glucocorticoid receptor and glycogen synthase were measured by real-time fluorescence quantitative PCR. Results the content of carnosine in gastrocnemius in group A was significantly higher than that in group C (p0.05) and group B was significantly higher than that in group C (P 0.01). The activity of hexokinase in gastrocnemius was significantly lower in group A than in group D. The liver glycogen content of rats in group C was significantly higher than that in group D (P 0.01). The mRNA expression of glucose-6-phosphatase in liver of rats in group B and C was significantly higher than that in group D (p0.01A) and the expression of glucocorticoid receptor (Glucocorticoid receptor) in liver of group D (p0.01A). The relative expression of glycogen synthase (mRNA) in liver of group C was significantly higher than that of group B (P 0.01). Conclusion: the relative expression of mRNA of insulin receptor in group A is significantly higher than that in group B (P 0.01). Conclusion: supplement of palanine can increase the expression of glycogen synthase in liver of rats. Promoting intermittent training of the use of blood glucose in rat muscles, Increased liver glycogen decomposition, medium dose effect is better.
