海洋芽孢桿菌中Iturin A分離鑒定研究性實驗教學(xué)探索

【摘要】:以一株海洋Bacillus sp.細(xì)菌為對象,進(jìn)行天然藥物化學(xué)研究性實驗的教學(xué)探索。通過優(yōu)化菌株發(fā)酵條件、改進(jìn)文獻(xiàn)提取分離純化方案,高效率地分離純化出目標(biāo)產(chǎn)物Iturin A,并指導(dǎo)學(xué)生綜合運(yùn)用質(zhì)譜,1HNMR結(jié)合文獻(xiàn)調(diào)研鑒定產(chǎn)物結(jié)構(gòu)。該實驗合理使用制備高效液相色譜儀,液質(zhì)聯(lián)用儀、核磁共振儀等大型儀器設(shè)備,改進(jìn)了傳統(tǒng)天然藥物化學(xué)實驗內(nèi)容,實現(xiàn)了學(xué)生實驗中微量天然產(chǎn)物快速、高效、可視化的分離與純化。實驗有助于激發(fā)學(xué)生學(xué)習(xí)興趣,使其全面深刻地認(rèn)識微生物天然產(chǎn)物的科學(xué)研究流程,了解掌握不同大型儀器的應(yīng)用范圍和操作技術(shù),提升操作技能和創(chuàng)新能力。
[Abstract]:With a marine Bacillus sp. Bacteria as the object, carry on the natural medicine chemistry research experiment teaching exploration. By optimizing the fermentation conditions of the strain, improving the extraction and purification scheme of the literature, the target product Iturin A was isolated and purified efficiently, and the students were instructed to use mass spectrometry and 1HNMR combined with literature investigation to identify the structure of the product. In this experiment, the preparation of high performance liquid chromatograph, liquid mass spectrometer, nuclear magnetic resonance instrument and other large instruments and equipment were reasonably used, which improved the content of traditional natural drug chemistry experiment and realized the rapid and high efficiency of trace natural products in students' experiment. Visual separation and purification. The experiment is helpful to stimulate students' interest in learning, to have a comprehensive and profound understanding of the scientific research process of microbial natural products, to master the application scope and operation technology of different large-scale instruments, and to improve their operation skills and innovative ability.
