綠茶多酚可能通過促進自噬發(fā)揮腎臟保護作用

【摘要】:目的探討綠茶多酚(GTPs)對高脂膳食大鼠腎臟自噬水平的影響。方法體質(zhì)量160~180g雄性Wistar大鼠30只,適應性喂養(yǎng)1w后,隨機分為正常對照組(CON)、高脂膳食組(HFD)和高脂膳食+綠茶多酚干預組(HFD+GTPs),共3組,每組10只。對照組和高脂膳食組分別自由攝食標準飼料和高脂膳食飼料,高脂膳食+綠茶多酚干預組在自由攝食高脂膳食的同時按照體質(zhì)量給予綠茶多酚(200mg/kg.bw)。18w后斷頭處死大鼠并收集全血及腎臟組織;用試劑盒測定甘油三酯(TG)、膽固醇、血糖和丙二醛(MDA)指標;免疫組化法測4-HNE水平;Western-blotting法分別測定Beclin-1、LC3Ⅱ/LC3Ⅰ和p62的蛋白表達水平。結(jié)果血膽固醇水平高脂膳食組高于對照組;腎皮質(zhì)MDA和4-HNE水平及Beclin-1、LC3Ⅱ/LC3Ⅰ和p62的蛋白表達水平高脂膳食+綠茶多酚干預組均高于對照組;提示綠茶多酚干預可以改善上述指標。結(jié)論高脂膳食會導致大鼠腎臟自噬受阻,氧化應激水平增高,綠茶多酚干預可增強腎臟自噬流,并降低氧化應激水平。
[Abstract]:Objective to investigate the effect of green tea polyphenols (GTPs) on renal autophagy in rats with high fat diet. Methods 30 male Wistar rats weighing 160 脳 180 g were randomly divided into three groups: normal control group (CON), high fat diet group (HFD) and high fat diet green tea polyphenols intervention group (HFD GTPs), with 10 rats in each group. The control group and the high-fat diet group were free to eat standard feed and high-fat diet feed, respectively. In the high fat diet green tea polyphenols intervention group, green tea polyphenols (200mg/kg.bw) were given according to body mass while eating high fat diet freely. After 18 weeks, the rats were decapitated and the whole blood and kidney tissue were collected.
