結(jié)核分枝桿菌臨床分離株利福平和異煙肼耐藥性檢測(cè)方法比較
[Abstract]:Objective to investigate the application value of gene chip method, modified Roche culture and proportional method in the determination of drug resistance of Mycobacterium tuberculosis (MTB) rifampicin (RFP) and isoniazid (INH). Methods the 6 loci of RFP resistance gene rpo B in 200 clinical isolates of MTB were detected by gene chip method. One locus of 13 mutants of kat G, two mutants and 15 positions of INH inh A gene promoter, and one C-T mutation were detected. At the same time, the MTB strains were cultured in Roche culture and tested by proportional method, and the sensitivity, specificity and coincidence rate of the results were compared. Results 152 susceptible strains, 48 drug-resistant strains, 161 sensitive strains and 39 drug-resistant strains were detected by microarray assay. Using the results of Roche scale method as the criterion, the sensitivity of gene chip assay for the detection of drug resistance of RFP was 85.0% and 97.5% respectively, the specificity and coincidence rates were 82.8% and 91.0%, respectively, and the sensitivity, specificity and coincidence rate for detection of drug resistance of INH were 82.8% and 91.0%, respectively. Conclusion the microarray method can be used to screen the drug-resistant strains of Mycobacterium tuberculosis, and the diagnostic rate can be improved by combined detection with the drug sensitivity test of tuberculous bacteria.
