氣相色譜質譜聯(lián)用法對運動員尿液中內(nèi)源性類固醇水平特征的研究

【摘要】:目的:建立氣相色譜-質譜聯(lián)用(GC-MS)同時測定尿液中6種內(nèi)源性類固醇激素水平的方法,對廣東省運動員體內(nèi)的類固醇激素代謝特征進行統(tǒng)計歸納和比較分析,為廣東省運動員尿液中類固醇激素的生物護照的建立提供數(shù)據(jù)支持。方法:選取1 474名廣東省運動員(包括814名男隊員,660名女隊員)在賽前訓練期間的尿樣,并對其中主要的6種內(nèi)源性類固醇進行酶解、提取及衍生化后采用氣質聯(lián)用(GC-MS)方法定量分析。分析采用HP-1MS色譜柱(17 m×0.2 mm,0.11μm),恒流模式,使用氦氣為載氣,流量為1 m L·min-1,進樣量:1μL;分流比:10∶1;進樣口溫度和EI離子源溫度分別為280℃和200℃;質譜SCAN模式定性確證,掃描范圍m/z 40~460;選擇離子模式(SIM)下定量分析。結果:廣東省女運動員尿液中6種類固醇激素濃度及睪酮/表睪酮比值平均值均顯著低于國外女運動員(P0.05),其中睪酮濃度平均值僅為國外女運動員的12.2%;廣東省男運動員尿液中雄酮、本膽烷醇酮、睪酮、5β-雄烷-3α,17β-二醇4種類固醇激素濃度及睪酮/表睪酮比值平均值均顯著低于國外男運動員(P0.05),而表睪酮濃度平均值明顯高于國外男運動員(P0.05),5α-雄烷-3α,17β-二醇濃度平均值與國外男運動員基本持平(P0.05)。廣東省男、女運動員尿液中類固醇5α-雄烷-3α,17β-二醇/5β-雄烷-3α,17β-二醇濃度比值則均顯著高于國外男、女運動員的水平(P0.05),雄酮/本膽烷醇酮濃度比值與國外運動員基本持平。結論:本研究選取6種內(nèi)源性類固醇激素進行氣質聯(lián)用分析,并通過與國內(nèi)外運動員相關代謝指標的對比分析,對廣東運動員訓練期間的尿液中類固醇代謝特征進行歸納,為廣東省運動員尿液內(nèi)源性類固醇指標生物護照的建立提供數(shù)據(jù)支持。
