陰離子交換色譜-熒光檢測法測定人體血清中酪氨酸及其代謝產(chǎn)物

【摘要】:建立了陰離子交換色譜-熒光檢測法(AEC-FLD)同時(shí)測定人體血清中的酪氨酸(Tyr)、對羥苯基乳酸(PHPLA)和對羥苯基丙酸(PHPA)的分析新方法。樣品經(jīng)乙腈處理沉淀蛋白,離心后取上清液直接進(jìn)樣檢測。采用IonPac AG21保護(hù)柱(50×2mm)、IonPac AS21分析柱(250×2mm),流動(dòng)相為4mmol/L NaOH溶液,流速為0.45mL/min,熒光檢測器的激發(fā)波長和發(fā)射波長分別為277nm和340nm。優(yōu)化條件下三種物質(zhì)能得到良好的分離和檢測。Tyr、PHPLA和PHPA的線性范圍分別為0.30~25mg/L、0.025~2mg/L和0.025~2mg/L,相關(guān)系數(shù)在0.9998~0.9999之間,檢出限(S/N=3)分別為0.024mg/L、0.020mg/L和0.019mg/L。三個(gè)水平的加標(biāo)回收率在72.6%~99.1%之間,其相對標(biāo)準(zhǔn)偏差均小于6.62%。該方法簡單、快速、準(zhǔn)確,可用于實(shí)際樣品的測定。
[Abstract]:A new method for the simultaneous determination of tyrosine tyrosine tyrosine, p-hydroxyphenyl lactic acid PHPLA and p-hydroxyphenyl propionate PHPA in human serum by anion exchange chromatography and fluorescence detection was established. The protein was precipitated by acetonitrile treatment. After centrifugation, the supernatant was directly injected into the sample. The IonPac AG21 protection column 50 脳 2mm Pac AS21 was used for the analysis of 250 脳 2mm Pac column. The mobile phase was 4 mmol / L NaOH solution, the mobile phase was 4 mmol / L NaOH solution, the mobile phase was 4 mmol / L NaOH solution. At a flow rate of 0.45 mL / min, the excitation and emission wavelengths of the fluorescence detector are 277nm and 340 nm respectively. Under the optimized conditions, the linear range of the separation and detection of the three substances is 0.30 ~ 25mg / L 0.025mg / L 0.025mg / L and 0.0252mg / L, respectively, and the correlation coefficient is between 0.9998 and 0.9999. The detection limit is 0.024 mg / L 0.020 mg / L and 0.019 mg / L. the recoveries of the three levels are between 72.6% and 99.1%, and the relative standard deviations are less than 6.62%. The method is simple, rapid and accurate, and can be used for the determination of practical samples.
