VEGF在家兔缺血心肌中的表達(dá)及其死后穩(wěn)定性研究

【摘要】:目的觀察心肌中VEGF在死后一定時(shí)間的穩(wěn)定性及表達(dá)的變化規(guī)律,評(píng)價(jià)其對(duì)死后診斷機(jī)體早期心肌缺血的作用。方法用冠狀動(dòng)脈結(jié)扎法建立家兔急性心肌缺血模型,應(yīng)用SP法染色、圖像分析和統(tǒng)計(jì)學(xué)處理系統(tǒng),檢測(cè)缺血心肌細(xì)胞內(nèi)VEGF陽(yáng)性表達(dá)面積和強(qiáng)度,并對(duì)VEGF在死后不同時(shí)間的穩(wěn)定性進(jìn)行比較。結(jié)果VEGF在缺血心肌組織中,見(jiàn)灶性或成片心肌細(xì)胞強(qiáng)陽(yáng)性表達(dá),部分細(xì)胞呈陰性或弱陽(yáng)性表達(dá),血管內(nèi)皮及平滑肌細(xì)胞表達(dá)弱陽(yáng)性,間質(zhì)表達(dá)陰性;在4℃放置,隨時(shí)間延長(zhǎng),缺血心肌組織VEGF陽(yáng)性表達(dá)有減弱趨勢(shì),至10d時(shí)缺血心肌組織仍可見(jiàn)VEGF在少許心肌細(xì)胞胞漿內(nèi)呈弱陽(yáng)性表達(dá)。在4℃放置10d以內(nèi),缺血心肌和正常心肌標(biāo)本之間VEGF陽(yáng)性表達(dá)具有極顯著性差異。結(jié)論VEGF能耐受一定程度自溶及腐敗的影響,其免疫組化檢測(cè)對(duì)于判斷死后4℃放置10d以內(nèi)的尸體是否出現(xiàn)過(guò)心肌缺血具有一定意義。
[Abstract]:Objective to observe the stability and expression of VEGF in myocardium at a certain time after death and to evaluate its role in the diagnosis of early myocardial ischemia. Methods A rabbit model of acute myocardial ischemia was established by coronary artery ligation. The area and intensity of VEGF positive expression in ischemic cardiomyocytes were detected by SP staining, image analysis and statistical processing system. The stability of VEGF at different time after death was compared. There was strong positive expression in focal or adult cardiomyocytes, negative or weak positive expression in some cells, weak positive expression in vascular endothelial and smooth muscle cells and negative expression in interstitial tissue. The positive expression of VEGF in ischemic myocardium tended to weaken, and VEGF was still weakly expressed in the cytoplasm of a few myocardial cells at 10 days after ischemia, and within 10 days at 4 鈩,
